入侵害蟲紅棕象甲核轉(zhuǎn)錄因子RfRelish編碼基因的克隆與表達(dá)特征分析

【摘要】:紅棕象甲R(shí)hynchophorus ferrugineus Olivier是我國(guó)的一種高風(fēng)險(xiǎn)性外來有害生物,是棕櫚植物的一種世界性毀滅害蟲。該害蟲危害隱蔽,田間種群已產(chǎn)生抗藥性而且生物防治的控制效率一直較低。因此,尋找針對(duì)該害蟲新的、有效的可持續(xù)控制策略迫在眉睫。昆蟲免疫是昆蟲對(duì)抗病原天敵的最主要途徑,然而對(duì)于該害蟲的免疫防御機(jī)理知之甚少。因此,本文通過紅棕象甲核轉(zhuǎn)錄因子RfRelish的克隆和定量表達(dá)分析等手段解析其潛在的免疫功能角色。研究發(fā)現(xiàn),該核轉(zhuǎn)錄因子RfRelish基因的全長(zhǎng)cDNA序列為2707 bp,開放閱讀框?yàn)?148 bp。其中,5'非編碼區(qū)為87 bp,3'非編碼區(qū)為472 bp,包含AAAAAAA加尾信號(hào)并含有長(zhǎng)為12bp的poly(A)尾。該基因編碼一個(gè)由714個(gè)氨基酸組成的蛋白質(zhì)。該蛋白質(zhì)的等電點(diǎn)為5.46,其分子量為81.5K Da。該編碼蛋白具有N末端的Rel homology domain(RHD)、IPT domain(Ig-like,plexins,transcription factors)和由四個(gè) ankyrin repeats組成的IκB-like domain等三個(gè)NF-κB家族蛋白所具有的典型保守結(jié)構(gòu)域。同時(shí),該蛋白具有核轉(zhuǎn)位定位信號(hào),該信號(hào)由第210-213位氨基酸殘基KRPR組成。這些保守結(jié)構(gòu)域的存在說明紅棕象甲R(shí)fRelish基因編碼的蛋白質(zhì)是一類具有重要功能的轉(zhuǎn)錄因子。熒光定量分析發(fā)現(xiàn),該基因在紅棕象甲不同組織中的表達(dá)量存在極顯著差異;其中,在免疫相關(guān)組織脂肪體、血淋巴、嗉囊和中腸中的表達(dá)量顯著高于非免疫相關(guān)組織。注射大腸桿菌后,紅棕象甲脂肪體中RfRelish基因的表達(dá)量顯著提高。而注射金黃色葡萄球菌6h后,僅紅棕象甲腸道中的RjtRelish基因表達(dá)量明顯高于對(duì)照組。已有證據(jù)顯示,紅棕象甲的腸道中棲居著豐富的細(xì)菌群落。因此,這說明RfRelish基因的編碼蛋白不僅在紅棕象甲的系統(tǒng)免疫中發(fā)揮重要作用,而且在腸道菌群穩(wěn)態(tài)的調(diào)控中也扮演關(guān)鍵角色。
[Abstract]:Red brown weevil (Rhynchophorus ferrugineus Olivier) is a high risk alien pest in China and a worldwide destructive pest of palm plants. The pest is concealed, the field population has developed resistance and the control efficiency of biological control has been low. Therefore, it is urgent to find a new and effective sustainable control strategy for the pest. Insect immunity is the most important way for insects to fight natural enemies, but little is known about the immune defense mechanism of insect pests. Therefore, the potential role of RfRelish was analyzed by cloning and quantitative expression analysis of its transcription factor. It was found that the full-length cDNA sequence of RfRelish gene was 2707 BP and the open reading frame was 2148 BP. Among them, the non-coding region of 5'is 87bp / 3'and the non-coding region is 472bp. it contains AAAAAAA signal with tail and 12bp's long polycaudal tail. The gene encodes a protein consisting of 714 amino acids. The isoelectric point of the protein is 5.46 and its molecular weight is 81.5 K Da. The encoding protein has the typical conserved domain of the N-terminal Rel homology domain Rel homology domain like Ig-like transcription factors and three NF- 魏 B family proteins, I 魏 B-like domain, composed of four ankyrin repeats. At the same time, the protein has nuclear translocation signal, which is composed of 210-213 amino acid residues KRPR. The existence of these conserved domains suggests that the proteins encoded by the RfRelish gene of the red brown weevil are a class of transcription factors with important functions. Fluorescence quantitative analysis showed that there were significant differences in the expression of the gene in different tissues of red brown weevil, and the expression of the gene was significantly higher in immune related tissues than in non-immune related tissues, such as fat body, hemolymph, crop and midgut. After the injection of E. coli, the expression of RfRelish gene in the fat body of red brown weevil was significantly increased. However, the expression of RjtRelish gene in the intestine of red brown weevil was significantly higher than that of the control group 6 h after injection of Staphylococcus aureus. There is evidence that red brown elephant beetles live in abundant bacterial communities in their intestines. Therefore, the encoding protein of RfRelish gene not only plays an important role in the systemic immunity of red brown weevil, but also plays a key role in the regulation of intestinal flora homeostasis.
