糯玉米種質(zhì)waxy基因的Eco-TILLING分析

【摘要】:本研究利用Eco-TILLING分析技術(shù)結(jié)合測(cè)序技術(shù),對(duì)糯玉米種質(zhì)材料的waxy基因序列變異進(jìn)行分析。結(jié)果顯示,在waxy基因從第9外顯子到第14外顯子總長為1 752 bp的序列中共發(fā)現(xiàn)72個(gè)突變位點(diǎn);我們將156份糯玉米材料劃分為13個(gè)單倍型,發(fā)現(xiàn)不同來源糯玉米種質(zhì)糯質(zhì)表現(xiàn)型的遺傳基礎(chǔ)表現(xiàn)出較大差異,在DNA序列上表現(xiàn)為SNP、InDel等不同類型的突變。對(duì)waxy基因序列進(jìn)行進(jìn)化分析表明,中國不同生態(tài)區(qū)糯玉米waxy基因序列具有不同的進(jìn)化來源和進(jìn)化方向,主要分為種群Ⅰ、種群Ⅱ兩個(gè)大的種群,其中種群Ⅱ?yàn)橹袊髂系貐^(qū)特有的糯質(zhì)玉米類型。研究探索了中國糯玉米種質(zhì)遺傳機(jī)制,發(fā)掘了有育種價(jià)值的有利等位基因資源,為糯玉米育種提供了理論依據(jù)和技術(shù)支持。
[Abstract]:In this study, Eco-TILLING analysis and sequencing were used to analyze the variation of waxy gene in waxy maize germplasm. The results showed that 72 mutation sites were found in the total length of 1 752bp from exon 9 to exon 14 of waxy gene, and we divided 156 waxy maize materials into 13 haplotypes. It was found that the genetic basis of waxy phenotypes of waxy maize germplasm from different sources was different, and the DNA sequence showed different types of mutations such as SNPN InDel. The evolutionary analysis of waxy gene sequence showed that the waxy gene sequence of waxy maize in different ecological regions of China had different evolutionary sources and evolutionary directions, mainly divided into two large populations: population 鈪,
