轉(zhuǎn)SERCA 2a基因?qū)毙孕姆款潉?dòng)模型心房肌細(xì)胞L型鈣通道影響的研究

【摘要】:目的:研究AAV9-SERCA 2a基因轉(zhuǎn)染對(duì)急性心房顫動(dòng)(房顫)動(dòng)物模型心房肌細(xì)胞上L型鈣通道表達(dá)的影響。方法:采用60只新西蘭大白兔為實(shí)驗(yàn)對(duì)象,采用心包注射法轉(zhuǎn)染AAV9-SERCA 2a基因;將實(shí)驗(yàn)對(duì)象隨機(jī)分為4組:房顫組、轉(zhuǎn)熒光蛋白房顫組、轉(zhuǎn)SERCA 2a基因房顫組和對(duì)照組。轉(zhuǎn)染后建立急性房顫模型,采用膜片鉗及熒光免疫組織化學(xué)法檢測(cè)心房肌細(xì)胞上L型鈣通道的電流變化及其L型鈣通道蛋白表達(dá)的情況。結(jié)果:轉(zhuǎn)SERCA 2a基因房顫組心房肌細(xì)胞上LVDCCa1c蛋白表達(dá)明顯高于房顫組(P0.05),與對(duì)照組比較差異無統(tǒng)計(jì)學(xué)意義。轉(zhuǎn)SERCA 2a基因房顫組心房肌細(xì)胞上L型鈣通道電流密度高于房顫組,差異有統(tǒng)計(jì)學(xué)意義(P0.05),與對(duì)照組比較差異無統(tǒng)計(jì)學(xué)意義。結(jié)論:轉(zhuǎn)SERCA 2a基因治療可減少急性房顫模型心房肌細(xì)胞鈣超載,減少房顫對(duì)L型鈣通道電流密度的影響,對(duì)防止心臟電重構(gòu)具有積極的意義。
[Abstract]:Aim: to study the effect of AAV9-SERCA 2a gene transfection on the expression of L-type calcium channel in atrial myocytes of acute atrial fibrillation (AF) animal model. Methods: sixty New Zealand white rabbits were used to transfect AAV9-SERCA 2a gene by pericardial injection. The subjects were randomly divided into 4 groups: atrial fibrillation group, fluorescent protein atrial fibrillation group, SERCA 2a gene atrial fibrillation group and control group. The acute atrial fibrillation model was established after transfection. The changes of L-type calcium channel current and the expression of L-type calcium channel protein in atrial myocytes were detected by patch clamp and fluorescence immunohistochemistry. Results: the expression of LVDCCa1c protein in atrial myocytes in SERCA 2a gene group was significantly higher than that in atrial fibrillation group (P 0.05). The L-type calcium channel current density in atrial myocytes in SERCA 2a gene group was significantly higher than that in atrial fibrillation group (P 0.05), but there was no significant difference compared with the control group. Conclusion: SERCA 2a gene therapy can reduce the calcium overload of atrial myocytes in acute atrial fibrillation model, reduce the effect of atrial fibrillation on L-type calcium channel current density, and have a positive significance in preventing cardiac electrical remodeling.
