日本鰻鱺TLR21基因的鑒定、免疫應(yīng)答與啟動(dòng)子分析

【摘要】:為研究TLR21(Toll like receptor 21)在低等脊椎動(dòng)物中的功能及表達(dá)調(diào)控機(jī)制,我們擴(kuò)增獲得了日本鰻鱺TLR21(Aj TLR21)c DNA序列,其編碼的蛋白具有TLR家族的共同特征。Aj TLR21基因結(jié)構(gòu)與其他魚類和兩棲類TLR21相同,由單個(gè)外顯子編碼。熒光定量結(jié)果顯示,Aj TLR21在血液、鰓、脾臟、中腎等11個(gè)組織/器官中轉(zhuǎn)錄表達(dá),其中在血液中表達(dá)量最高。經(jīng)Poly I:C誘導(dǎo)后8h,Aj TLR21在脾臟和中腎中的表達(dá)量顯著性上調(diào);誘導(dǎo)后16h,Aj TLR21在血液、鰓、腸和脾臟中的表達(dá)量顯著性上調(diào)(P0.05)。雙熒光素酶報(bào)告基因結(jié)果顯示,在Aj TLR21 5′上游調(diào)控序列 1179 bp到+117 bp存在Poly I:C調(diào)節(jié)的正調(diào)控元件。經(jīng)Edwardsiella tarda誘導(dǎo)后16h和72h,Aj TLR21分別在血液和中腎組織的表達(dá)量顯著性上調(diào),表明Aj TLR21同時(shí)也參與了抗細(xì)菌免疫應(yīng)答,其在機(jī)體免疫系統(tǒng)中的功能具有多樣性。研究對于理解日本鰻鱺Aj TLR21的免疫學(xué)功能具有重要的理論意義和應(yīng)用價(jià)值。
[Abstract]:In order to study the function and expression regulation mechanism of TLR21(Toll like receptor 21 in low vertebrate, we amplified the TLR21(Aj TLR21)c DNA sequence of Japanese eel. The protein encoded by Aj TLR21 has the same structure as other fish and amphibian TLR21, and is encoded by a single exon. Fluorescence quantitative analysis showed that Aj TLR21 was transcribed in 11 tissues / organs such as blood, Gill, spleen and mesonephrosis, and the highest expression was found in blood. The expression of Aj TLR21 was significantly up-regulated in spleen and midkidney at 8 h after induction by Poly I: C, and significantly up-regulated in blood, Gill, intestine and spleen at 16 h after induction. The results of double luciferase reporter gene showed that there were positive regulatory elements of Poly I: C in the upstream regulatory sequence of Aj TLR21 5'from 1179 BP to 117bp. The expression of Aj TLR21 was significantly up-regulated in blood and midkidney tissues at 16 h and 72 h after induction by Edwardsiella tarda, indicating that Aj TLR21 also participated in the anti-bacterial immune response, and its function in the immune system was diverse. The study has important theoretical significance and application value for understanding the immunological function of Japanese eel Aj TLR21.
