枳SnRK2.4和SnRK2.6轉(zhuǎn)化檸檬及轉(zhuǎn)基因植株多胺含量與抗性分析

【摘要】:SnRK2(Sucrose non-fermenting 1-related protein kinase2,蔗糖非酵解型蛋白激酶2)是植物中特有的一類絲氨酸/蘇氨酸蛋白激酶,在ABA信號途徑及植物抵御脅迫中起著關(guān)鍵作用。多胺是直接參與調(diào)控植物抗逆性的信號分子,其合成受到ABA信號途徑的調(diào)控。但是SnRK2在調(diào)控多胺合成中的作用尚不清楚。本研究克隆了枳(Poncirus trifoliata(L.)Raf.)兩個SnRK2基因PtrSnRK2.4與PtrSnRK2.6,利用根癌農(nóng)桿菌介導檸檬轉(zhuǎn)化獲得了轉(zhuǎn)基因植株,并對PtrSnRK2.4檸檬轉(zhuǎn)基因系多胺含量及抗性進行分析。主要研究結(jié)果如下:1、從枳中克隆出PtrSnRK2.4,全長1026 bp,編碼341個氨基酸;PtrSnRK2.6,全長1092 bp,編碼363個氨基酸。PtrSnRK2.4和PtrSnRK2.6分別屬于GroupⅢ和GroupⅡ家族。2、利用根癌農(nóng)桿菌介導轉(zhuǎn)化檸檬上胚軸,共獲得PtrSnRK2.4轉(zhuǎn)基因檸檬24株,PtrSnRk2.6轉(zhuǎn)基因檸檬8株。對PtrSnRK2.4轉(zhuǎn)基因系與PtrSnRK2.6轉(zhuǎn)基因系相關(guān)基因表達量檢測,發(fā)現(xiàn)在轉(zhuǎn)基因株系中ABF1、ABF2和ADC表達量均高于野生型,PtrSnRK2.6轉(zhuǎn)基因株系中ABF4也上調(diào)表達。并且PtrSnRK2.4轉(zhuǎn)基因植株中腐胺含量均高于野生型。3、對PtrSnRK2.4轉(zhuǎn)基因植株抗性進行分析,脫水處理50 min后,野生型比轉(zhuǎn)基因植株萎蔫更嚴重,與之一致的是前者相對失水率和電導率也高于后者。表明PtrSnRK2.4轉(zhuǎn)基因植株比野生型表現(xiàn)出更高的抗旱能力。
[Abstract]:SnRK2(Sucrose non-fermenting 1-related protein kinase2 (sucrose non-fermentable protein kinase 2) is a unique serine / threonine protein kinase in plants, which plays a key role in ABA signaling pathway and plant resistance to stress. Polyamines are signaling molecules directly involved in the regulation of plant stress resistance, and their synthesis is regulated by the ABA signaling pathway. However, the role of SnRK2 in the regulation of polyamine synthesis is unclear. In this study, Poncirus trifoliata L. Raf. was cloned. Two SnRK2 genes, PtrSnRK2.4 and PtrSnRK 2.6, were transformed into lemon by Agrobacterium tumefaciens. The polyamines content and resistance of PtrSnRK2.4 lemon transgenic lines were analyzed. The main results were as follows: 1. PtrSnRK2.4 was cloned from trifoliate orange, with a total length of 1026 BP, encoding 341 amino acids PtrSnRK2.6. the total length of PtrSnRK2.6 was 1092 BP, and the encoding of 363-amino acid. PtrSnRK2.4 and PtrSnRK2.6 belonged to Group 鈪,
