廣西1例HIV-1G亞型毒株全長(zhǎng)基因序列的測(cè)定和分析

【摘要】:目的對(duì)廣西1例HIV-1 G亞型毒株感染者體內(nèi)病毒近似全長(zhǎng)基因組進(jìn)行序列測(cè)定并進(jìn)行系統(tǒng)進(jìn)化分析。方法對(duì)前期鑒定的1例HIV-1G亞型毒株感染者進(jìn)行背景信息收集,并采集其外周血,提取病毒RNA,利用逆轉(zhuǎn)錄巢式-PCR技術(shù)分兩段擴(kuò)增并測(cè)定病毒近似全長(zhǎng)基因序列。利用MEGA6軟件對(duì)全長(zhǎng)序列進(jìn)行系統(tǒng)進(jìn)化分析。結(jié)果獲得了病毒近似全長(zhǎng)基因組序列,長(zhǎng)度為8847 bp。在系統(tǒng)進(jìn)化樹中,該毒株與G亞型參考株成簇,Bootstrap值為100%。系統(tǒng)溯源分析顯示,該毒株與廣西首次報(bào)道的G亞型JN106043毒株關(guān)系最近(Bootstrap值為100%,基因離散率為5%)。結(jié)論該毒株可能是由廣西毒株傳播所致,提示G亞型毒株在廣西地區(qū)可能存在一定的局部流行,需進(jìn)一步監(jiān)測(cè)。該毒株序列的測(cè)定和分析對(duì)于廣西地區(qū)HIV流行狀況分析具有重要意義。
[Abstract]:Objective to sequence and analyze the approximate full-length genome of a case of HIV-1 G subtype infection in Guangxi. Methods the background information was collected from a case of HIV-1G subtype infection, and the peripheral blood was collected, virus RNAs were extracted, and the approximate full-length gene sequence of the virus was amplified and sequenced by reverse transcription-nested polymerase chain reaction (RT-PCR). The phylogenetic analysis of full-length sequences was carried out by using MEGA6 software. Results the approximate full-length genome sequence of the virus was obtained, the length of which was 8847 BP. In phylogenetic tree, the Bootstrap value of this strain and G subtype reference strain is 100. Systematic traceability analysis showed that the relationship between this strain and the G subtype JN106043 strain reported for the first time in Guangxi was the closest bootstrap value was 100 and the gene dispersion rate was 5%. Conclusion this strain may be caused by the spread of Guangxi strain, suggesting that there may be a local epidemic of subtype G strain in Guangxi, which needs further monitoring. The determination and analysis of the sequence of the strain is of great significance for the analysis of the epidemic status of HIV in Guangxi.
