【摘要】：對豬瘟病毒(CSFV)、豬繁殖與呼吸障礙綜合征病毒(PRRSV)和豬流行性乙型腦炎病毒(JEV)聯(lián)合共檢基因芯片的初步應(yīng)用進行了研究。選用夾縫針,BaiOR點樣緩沖液,確定靶基因質(zhì)量濃度為200mg/L,各樣點中心間距300μm,樣點直徑100μm,在相對濕度為50%~60%、8~30℃條件下用晶芯?SmartArrayerTM48點樣儀在氨基化基片上接觸式點樣,成功制備了CSFV-PRRSV-JEV檢測基因芯片。將標記后的探針基因與PRRSV-CSFV-JEV基因芯片經(jīng)預(yù)雜交、雜交、洗滌干燥后,用晶芯?LuxScanTM10K微陣列芯片掃描儀掃描觀察結(jié)果。結(jié)果表明:制備的芯片特異性強,檢測靈敏度可達0.295μg/L,4℃條件下可保存120d;用制備的芯片對臨床67份疑似繁殖障礙性疾病的病料進行檢測,與RT-PCR檢測結(jié)果符合率均在94%以上,為豬繁殖障礙性疾病的檢測提供了理想的檢測方法。
