冷藏期間蘋(píng)果果實(shí)能量和NADP-蘋(píng)果酸酶及基因表達(dá)的變化

【摘要】:為研究冷藏期間蘋(píng)果果實(shí)能量水平和NADP-蘋(píng)果酸酶(NADP-ME)及基因表達(dá)的變化情況,本研究以‘長(zhǎng)富2’蘋(píng)果果實(shí)為實(shí)驗(yàn)材料,定期測(cè)定果實(shí)硬度、可溶性固形物等品質(zhì)指標(biāo)和能量相關(guān)物質(zhì)三磷酸腺苷(ATP)、二磷酸腺苷(ADP)、磷酸腺苷(AMP)含量和能荷變化及NADP-ME、琥珀酸脫氫酶(SDH)活性。結(jié)果顯示,冷藏期間,‘長(zhǎng)富2’果實(shí)ATP含量貯藏前60 d下降,而后恢復(fù)到貯藏初期水平,ADP和AMP貯藏前60 d稍有降低后,隨時(shí)間延長(zhǎng)顯著增加;NADP-ME和琥珀酸脫氫酶(SDH)活性均隨貯藏時(shí)間延長(zhǎng)逐漸降低;輔酶Ⅱ(NADP+)和還原型輔酶Ⅱ(NADPH)含量在貯藏前120 d先增加后下降,與能荷變化相似;Mdcy ME1-Mdcy ME3基因表達(dá)雖在時(shí)間上有一定差異,但表現(xiàn)出相似的先增加后下降的趨勢(shì)。冷藏期間蘋(píng)果果實(shí)能量水平和利用效率發(fā)生了改變,且能量水平與NADP-ME活性及其基因表達(dá)在統(tǒng)計(jì)學(xué)上存在相關(guān)性。
[Abstract]:In order to study the changes of energy level, NADP- malate enzyme NADP-MEE and gene expression in apple fruit during cold storage, the fruit of 'Changfu 2'apple was used as experimental material, and the firmness of fruit was measured regularly. The content and energy charge of adenosine triphosphate (ATP), adenosine diphosphate (ADPN), adenosine monophosphate (AMPP), and the activities of NADP-MES and succinate dehydrogenase (SDH). The results showed that the ATP content of 'Changfu 2' fruit decreased 60 days before storage, and then returned to the initial storage level 60 days before storage, and then decreased slightly 60 days before storage.
