玉米ZmBRI1基因的克

【摘要】:油菜素內(nèi)酯(BRs)是一種重要的甾族類(lèi)激素,在植物的生長(zhǎng)過(guò)程中起著重要的作用。本研究利用同源克隆的方法,從玉米B73自交系中獲得了一個(gè)新的油菜素內(nèi)酯受體蛋白編碼基因ZmBRI1,該基因全長(zhǎng)為3369 bp,編碼1122個(gè)氨基酸。亞細(xì)胞定位分析表明,ZmBRI1蛋白定位于細(xì)胞膜上,而且ZmBRI1在各個(gè)玉米的各個(gè)組織器官中都有表達(dá),其中在幼嫩的組織中表達(dá)最高。利用轉(zhuǎn)基因技術(shù)將ZmBRI1導(dǎo)入BR不敏感突變體bri1-5中,獲得的轉(zhuǎn)基因植株修復(fù)了其表型,特別是植株高度、葉片形態(tài)和果莢大小。與突變體bri1-5比較,油菜素內(nèi)酯(brassinolide,BL)處理顯著抑制轉(zhuǎn)基因株系根系生長(zhǎng);丙環(huán)唑(propiconazole,Pcz)處理顯著抑制了下胚軸生長(zhǎng);同時(shí)轉(zhuǎn)基因株系DWF4和CPD基因的表達(dá)量下降。此外,在野生型(Ws)中過(guò)表達(dá)ZmBRI1,顯著提高了擬南芥在ABA抑制條件下的種子萌發(fā)率和植株生長(zhǎng),下調(diào)了ABA響應(yīng)基因RD29A、RD29B、ABI5和RAB18的表達(dá)。因此,ZmBRI1不僅參與了植物的形態(tài)建成和BR的信號(hào)傳導(dǎo),而且參與調(diào)控了植物對(duì)ABA信號(hào)的響應(yīng)。
[Abstract]:Brassinolide (BRs) is an important steroid hormone, which plays an important role in plant growth. In this study, a novel rapesinolactone receptor protein encoding gene ZmBRI1 was obtained from maize B73 inbred line by homologous cloning. The gene is 3369 BP and encodes 1122 amino acids. Subcellular localization analysis showed that ZmBRI1 protein was located on the cell membrane, and ZmBRI1 was expressed in all tissues and organs of maize, and the highest expression was found in young tissues. ZmBRI1 was introduced into Br insensitive mutant bri1-5 by transgenic technique. The transgenic plants repaired their phenotypes, especially plant height, leaf morphology and pod size. Compared with the mutant bri1-5, Brassinolide (BLL) treatment significantly inhibited the root growth of transgenic lines, the propiconazoleoledol (Pcz) treatment significantly inhibited the Hypocotyl growth, and the expression of DWF4 and CPD genes decreased in the transgenic lines. In addition, the overexpression of ZmBRI1 in wild type Ws1 significantly increased the seed germination rate and plant growth of Arabidopsis thaliana under the condition of ABA inhibition, and down-regulated the expression of ABA response gene RD29A, RD29B, ABI5 and RAB18 in Arabidopsis thaliana. ZmBRI1 is involved not only in plant morphogenesis and Br signal transduction, but also in plant response to ABA signal.
