佐太和HgS對(duì)藥物代謝酶和轉(zhuǎn)運(yùn)體基因表達(dá)的影響

【摘要】:藏藥佐太(主要含β-HgS)和中藥朱砂(96%HgS)是傳統(tǒng)醫(yī)藥中的配伍成分,其含重金屬(汞)的安全問(wèn)題和中西藥合用時(shí)的相互作用問(wèn)題日益受到關(guān)注。本實(shí)驗(yàn)就其對(duì)藥物代謝基因的影響進(jìn)行了研究。小鼠連續(xù)7 d口服給予不同劑量佐太、HgS(10,30,100,300 mg·kg-1),或氯化汞(Hg Cl2,33.6 mg·kg-1),RT-PCR檢測(cè)藥物代謝酶和轉(zhuǎn)運(yùn)體蛋白相關(guān)基因表達(dá)。佐太和HgS對(duì)Ⅰ相(CYP1A2,CYP2B10,CYP3A11,CYP4A10)和Ⅱ相(UGT1A1,UGT2A3,SULT1A1,SULT2A1)藥物代謝酶以及轉(zhuǎn)運(yùn)體蛋白(OATP1A1,OATP1A4,OATP1B2,OATP2B1,MRP1,MRP2,MRP3,MRP4)均無(wú)明顯影響,而Hg Cl2顯著上調(diào)CYP2B10,CYP4A10,UGT1A1,UGT2A3,SULT1A1,SULT2A1,OATP1A4,MRP1,MRP3,MRP4,對(duì)OATP1A1有抑制作用。綜上,佐太和HgS在300 mg·kg-1的劑量下對(duì)肝臟代謝基因的表達(dá)沒(méi)有產(chǎn)生明顯的影響,而1/10等汞量的Hg Cl2則明顯上調(diào)或改變Ⅰ相、Ⅱ相藥物代謝基因和轉(zhuǎn)運(yùn)體基因的表達(dá)。
[Abstract]:Zo Tai (mainly containing 尾 -HgS) and Chinese medicine cinnabar 96 HgS are the compatible components of traditional medicine. The safety of heavy metals (mercury) and the interaction between Chinese and western medicine have been paid more and more attention. In this study, the effect of drug metabolism gene was studied.
