雛鴨坦布蘇病毒分離鑒定及全基因序列分析

【摘要】:山東濰坊地區(qū)某鴨場(chǎng)養(yǎng)殖的21日齡櫻桃谷鴨突然發(fā)病,病鴨主要表現(xiàn)翅腿癱瘓等神經(jīng)癥狀,排黃綠色或黃白色稀糞,病死率5%左右。為了確定發(fā)病鴨群感染病原的種類(lèi),首先采集病死鴨肝臟,處理后接種9日齡~11日齡SPF雞胚進(jìn)行病原分離。其次,對(duì)分離的病原RT-PCR擴(kuò)增和序列測(cè)定,MEGA5繪制系統(tǒng)發(fā)育進(jìn)化樹(shù),DNA Star軟件分析核苷酸同源性。結(jié)果表明,通過(guò)SPF雞胚分離到一株坦布蘇病毒(TMUV),命名為T(mén)MUV-SDWF。該病毒株基因組全長(zhǎng)為10 990nt,編碼1個(gè)含3 426個(gè)氨基酸的多聚蛋白。系統(tǒng)發(fā)育進(jìn)化樹(shù)分析顯示,TMUV-SDAG毒株屬于Ⅰa分支。核苷酸同源性分析表明,TMUV-SDSG與GenBank上公布的16株不同TMUV分離株核苷酸同源性高達(dá)97.0%~99.8%。成功分離鑒定一株鴨坦布蘇病毒并對(duì)其進(jìn)行了遺傳進(jìn)化分析,為鴨坦布蘇病毒弱毒或滅活疫苗的篩選制備及該病毒分子流行病學(xué)的研究奠定了基礎(chǔ)。
[Abstract]:A 21-day-old Cherry Valley duck bred in Weifang area of Shandong Province suddenly developed symptoms such as paralysis of wings and legs and discharge of yellowish green or yellowish-white feces with a fatality rate of about 5%. In order to determine the pathogen of the infected duck, the dead duck liver was collected and inoculated with SPF embryos at 9 and 11 days of age to isolate the pathogen. Secondly, nucleotide homology was analyzed by Star software of phylogenetic tree derived from RT-PCR amplification and sequencing of isolated pathogens. The results showed that a strain of Tambusuvirus was isolated from chicken embryo of SPF and named TMUV-SDWF. The genome length of the virus strain is 10 990 nt, which encodes a polypeptide containing 3 426 amino acids. Phylogenetic tree analysis showed that the TMUV-SDAG strain belonged to branch 鈪,
