【摘要】：目的:采用免疫組化法觀察研究Sox2和Cdx2基因編碼蛋白在胃癌及其癌前病變組織中的表達(dá)并分析其與胃癌臨床病理生物學(xué)行為的關(guān)系,探討Sox2、Cdx2異常表達(dá)在胃癌發(fā)生發(fā)展中的作用機(jī)制及相互關(guān)系。方法:收集中國醫(yī)科大學(xué)附屬第一醫(yī)院腫瘤外科手術(shù)切除胃癌標(biāo)本257例,采用Envision免疫組織化學(xué)染色方法和HID-ABp H2.5-PAS黏蛋白組織化學(xué)染色法,觀察Sox2和Cdx2在胃癌及其癌前病變中的表達(dá),分析其表達(dá)的相互關(guān)系。結(jié)果:胃癌組織中Sox2的陽性表達(dá)率(64.98%)顯著低于正常胃黏膜(89.02%)、腸上皮化生(76.38%)和異型增生(71.01%)中的陽性表達(dá)率,P均0.05;彌漫型胃癌(71.54%)中的表達(dá)顯著高于腸型胃癌( 58. 25% ) ,P 0. 05。Cdx2 蛋白在胃癌組織中的陽性表達(dá)率( 53. 93% ) 明顯高于正常胃黏膜中的陽性表達(dá)率( 6. 74% ) ,在腸上皮化生組織中的陽性表達(dá)率( 88. 89% ) 顯著高于胃癌組織的陽性表達(dá)率,腸型胃癌中( 68. 75% ) 的表達(dá)水平高于彌漫型胃癌( 40. 38% ) ,P 均 0. 05。結(jié)論: Sox2 對胃表型的維持起重要作用,可能與彌漫型胃癌的發(fā)生關(guān)系更為密切。Cdx2 對腸表型的維持起重要作用,可能與腸型胃癌的發(fā)生更為密切。中的表達(dá)顯著高于腸型胃癌(58.25%),P0.05。Cdx2蛋白在胃癌組織中的陽性表達(dá)率(53.93%)明顯高于正常胃黏膜中的陽性表達(dá)率(6.74%),在腸上皮化生組織中的陽性表達(dá)率(88.89%)顯著高于胃癌組織的陽性表達(dá)率,腸型胃癌中(68.75%)的表達(dá)水平高于彌漫型胃癌(40.38%),P均0.05。結(jié)論:Sox2對胃表型的維持起重要作用,可能與彌漫型胃癌的發(fā)生關(guān)系更為密切。Cdx2對腸表型的維持起重要作用,可能與腸型胃癌的發(fā)生更為密切。
[Abstract]:Objective: to study the expression of Sox2 and Cdx2 gene coding proteins in gastric carcinoma and its precancerous lesions by immunohistochemical method, and to analyze the relationship between the expression and clinicopathologic behavior of gastric cancer. To investigate the mechanism and relationship of abnormal expression of Sox2 and Cdx2 in gastric carcinogenesis. Methods: a total of 257 gastric cancer specimens were collected from the first affiliated Hospital of China Medical University. Envision immunohistochemical staining and HID-ABp H2.5-PAS mucin histochemical staining were used.
