大豆GmDAO1基因的功能預(yù)測及表達(dá)分析

【摘要】:本研究通過在大豆基因組數(shù)據(jù)庫中檢索擬南芥AtDAO1在大豆中的同源基因,獲得了GmDAO1基因序列。通過對GmDAO1基因編碼的氨基酸序列及啟動子序列進行生物信息學(xué)分析,我們發(fā)現(xiàn)GmDAO1基因CDS序列全長951bp,編碼316個氨基酸。GmDAO1編碼的蛋白為親水性蛋白,具有1個N-糖基化位點、3個激酶磷酸化位點與1個豆蔻;稽c。結(jié)構(gòu)域分析表明GmDAO1含有雙加氧酶與2OG-Fe(II)加氧酶結(jié)構(gòu)域,是2-酮戊二酸依賴性雙加氧酶基因(2-ODD)家族的成員。GmDAO1預(yù)測的啟動子區(qū)域含有與激素、脅迫、光應(yīng)答、生物鐘調(diào)控和轉(zhuǎn)錄因子結(jié)合相關(guān)的順式作用元件。系統(tǒng)進化分析結(jié)果表明DAO1在豆科植物進化過程中比較保守。組織特異性表達(dá)分析結(jié)果顯示GmDAO1在葉片中表達(dá)量最低,在根中表達(dá)量最高。因此我們推測其可能參與生長素的代謝途徑。
[Abstract]:In this study, the GmDAO1 gene sequence was obtained by retrieving the homologous genes of Arabidopsis AtDAO1 in soybean genome database. By bioinformatics analysis of the amino acid sequence and promoter sequence encoded by GmDAO1 gene, we found that the GmDAO1 gene CDS sequence was all long 951bp and encoded 316 amino acid.GmDAO1 encoded eggs. White is hydrophilic protein with 1 N- glycosylation sites, 3 kinase phosphorylation sites and 1 myrisylation sites. Domain analysis shows that GmDAO1 contains dioxygenase and 2OG-Fe (II) oxygenase domain, and the promoter region predicted by 2- ketopamyl diacid dependent dioxygenase gene (2-ODD) gene promoter region contains hormone and stress, The results of phylogenetic analysis show that DAO1 is more conservative in the evolution of leguminous plants. The results of tissue specific expression analysis show that GmDAO1 has the lowest expression in the leaves and the highest expression in the root. Therefore, we speculate that it may be involved in the generation of auxin. Thank you for the way.
