【摘要】：研究應(yīng)用RNA-seq和聚合酶鏈?zhǔn)椒磻?yīng)確定了一個(gè)熱休克同源蛋白70-2(MsHSC70-2)基因以及應(yīng)用熒光定量PCR檢測(cè)紫花苜蓿不同季節(jié)和不同器官中MsHSC70-2的mRNA含量。結(jié)果表明:該基因的mRNA序列含有2 025bp,開(kāi)放閱讀框1 950bp,編碼649個(gè)氨基酸,該基因的蛋白分子量約70.97kDa,等電點(diǎn)4.74。同一器官不同季節(jié)MsHSC70-2mRNA含量不同(P0.05),夏季時(shí)不同器官間含量存在差異(P0.01),該基因在根、莖、葉和頂芽中的表達(dá)趨勢(shì)從四月份到九月份都是先升高后降低,表明MsHSC70-2的表達(dá)受日照長(zhǎng)度和溫度的調(diào)節(jié),其表達(dá)量變化影響了各器官的生長(zhǎng)發(fā)育,高表達(dá)促進(jìn)苜蓿的生長(zhǎng)發(fā)育,低表達(dá)抑制苜蓿的生長(zhǎng)發(fā)育,該基因表達(dá)的降低可能參與秋眠型苜蓿秋眠的發(fā)生。研究結(jié)果為進(jìn)一步研究紫花苜蓿HSC70-2的功能提供了依據(jù)。
[Abstract]:A heat shock homologous protein 70-2MsHSC70-2) gene was determined by RNA-seq and polymerase chain reaction. The mRNA content of MsHSC70-2 in different seasons and organs of alfalfa was detected by fluorescence quantitative PCR. The results showed that the mRNA sequence of the gene contained 2 025 BP, open reading frame 190 BP, encoding 649 amino acids, the molecular weight of the protein was about 70.97 kDa, and the isoelectric point was 4.74. The content of MsHSC70-2mRNA was different in different seasons of the same organ, and the content of P0.01 was different among different organs in summer. The expression trend of this gene in root, stem, leaf and terminal bud increased first and then decreased from April to September. The results showed that the expression of MsHSC70-2 was regulated by sunshine length and temperature. The changes of MsHSC70-2 expression affected the growth and development of various organs, the high expression promoted the growth and development of alfalfa, and the low expression inhibited the growth and development of alfalfa. The decrease of the gene expression may be involved in the autumn dormancy of alfalfa. The results provided a basis for further study on the function of alfalfa HSC70-2.
