四紋豆象小分子量熱激蛋白基因的鑒定與表達分析

【摘要】:本研究利用生物信息學方法對NCBI數(shù)據(jù)庫中四紋豆象(Callosobruchus maculatus)全部熱激蛋白進行分析,從中鑒定出一種中央?yún)^(qū)具有典型α-晶體蛋白質(zhì)結(jié)構(gòu)域的小分子量熱激蛋白基因,shsp27(Gen Bank編號FK669241.1)。采用實時熒光定量PCR(RT-q PCR)檢測了四紋豆象4齡幼蟲經(jīng)不同溫度(4、15、30、37℃以及4℃低溫處理24 h后回復(fù)30℃)脅迫處理24 h后,shsp27 m RNA的相對表達量。分析結(jié)果表明,在4齡四紋豆象幼蟲中,低溫(4、15℃)與高溫(37℃)脅迫處理后shsp27的相對表達量相對于對照組(30℃恒溫培養(yǎng)組)均有上調(diào)響應(yīng),但對高溫脅迫的響應(yīng)程度略強,低溫回復(fù)處理組的相對表達量也略高于對照組,但上升不顯著,初步認為shsp27基因表達與四紋豆象耐寒熱的適應(yīng)性無直接相關(guān)性。
[Abstract]:In this study, all heat shock proteins of Callosobruchus maculatus in NCBI database were analyzed by bioinformatics method, and a small molecular weight heat shock protein gene with typical 偽 -crystal protein domain in the central region was identified by Bank number FK669241.1.
