【摘要】:Virp1基因是第一個被報道可以結(jié)合RNA的Bromodomain(BRD)蛋白,在馬鈴薯紡錘塊莖類病毒(potato spindle tuber viroid,PSTVd)對宿主的感染及其復(fù)制中起重要作用。本研究根據(jù)NCBI登錄的virp1基因序列和桑樹基因組數(shù)據(jù)庫設(shè)計引物,進行RT-PCR擴增和克隆測序,獲得桑樹的同源MaGte4基因長1 771 bp的序列。生物信息學分析表明,基因有3個外顯子,CDS長1 683 bp,編碼560個氨基酸,有與乙酰化組蛋白賴氨酸具有特異相互作用的BRD及其extra-terminal(BET)結(jié)構(gòu)域,且該結(jié)構(gòu)及其氨基酸組成在各物種間比較保守,桑樹MaGte4具有和virp1蛋白非常相似的C-末端區(qū)域,RT-PCR分析基因在正常和花葉型萎縮病桑樹葉片、葉柄和葉脈中均有表達,推測MaGte4的功能可能和Virp1相類似。
[Abstract]:Virp1 gene is the first reported protein that can bind to RNA. It plays an important role in host infection and replication of potato spindle tuber viroid virus. In this study, primers were designed based on the virp1 gene sequence registered by NCBI and the mulberry genome database. The homologous MaGte4 gene of mulberry was amplified by RT-PCR and cloned and sequenced. The homologous MaGte4 gene of mulberry was 1 771bp in length. Bioinformatics analysis showed that three exons of the gene had a length of 1 683 BP, encoding 560 amino acids, and the BRD and its extra-terminalBETs had a specific interaction with acetylated histone lysine. Moreover, the structure and amino acid composition of mulberry were conserved among species. RT-PCR analysis of the C-terminal region of mulberry MaGte4, which is very similar to virp1 protein, was expressed in the leaves, petioles and veins of mulberry with normal and mosaic atrophy. It is speculated that the function of MaGte4 may be similar to that of Virp1.
