過表達(dá)GmATG8c基因提高大豆的低氮耐受性和產(chǎn)量

【摘要】:大豆是一種高需氮素作物,其籽粒發(fā)育所需氮素在很大程度上來源于"源"器官中氮素的再動(dòng)員,而細(xì)胞自噬是植物進(jìn)行氮素再動(dòng)員的主要形式之一。為了研究大豆細(xì)胞自噬關(guān)鍵基因GmATG8c的功能,本文利用35S啟動(dòng)子驅(qū)動(dòng)GmATG8c基因在大豆中過表達(dá)。通過農(nóng)桿菌介導(dǎo)的大豆子葉節(jié)轉(zhuǎn)化法獲得了3個(gè)獨(dú)立的單拷貝35S:GmATG8c轉(zhuǎn)基因大豆株系,半定量RT-PCR結(jié)果顯示,所有轉(zhuǎn)基因株系中目的基因均得到了高效表達(dá)。進(jìn)一步對(duì)轉(zhuǎn)基因大豆株系進(jìn)行的低氮耐受性和產(chǎn)量相關(guān)性狀分析結(jié)果表明,在低氮培養(yǎng)條件下,35S:GmATG8c轉(zhuǎn)基因大豆的生長(zhǎng)速率加快,二級(jí)分枝數(shù)和單株粒數(shù)增多,百粒重增加,單株產(chǎn)量顯著提高。說明GmATG8c是一個(gè)具有提高NUE(nitrogen use effi ciency)育種潛力的基因。
[Abstract]:Soybean is a kind of high nitrogen demand crop. The nitrogen needed for grain development of soybean comes from nitrogen remobilization in "source" organs to a great extent, and cellular autophagy is one of the main forms of nitrogen remobilization in plants. In order to study the function of key gene GmATG8c in soybean cell autophagy, 35s promoter was used to drive the overexpression of GmATG8c gene in soybean. Three independent single-copy 35S:GmATG8c transgenic soybean lines were obtained by Agrobacterium tumefaciens mediated cotyledon node transformation. The semi-quantitative RT-PCR results showed that the target genes were highly expressed in all transgenic lines. The results of analysis on low nitrogen tolerance and yield related characters of transgenic soybean lines showed that under low nitrogen culture, the growth rate of transgenic soybean was accelerated, the number of secondary branches and grains per plant increased, and the weight of 100 seeds increased. The yield per plant increased significantly. It is suggested that GmATG8c is a gene with the potential to improve NUE(nitrogen use effi breeding.
