基于線粒體COI和Cytb基因的中國灰蝶三亞科主要類群間的系統(tǒng)發(fā)育關(guān)系分析

【摘要】:【目的】針對中國灰蝶科中親緣關(guān)系較近的3個主要亞科[灰蝶亞科(Lycaeninae)、線灰蝶亞科(Theclinae)以及眼灰蝶亞科(Polyommatina)],基于線粒體基因序列數(shù)據(jù)研究它們主要類群間的系統(tǒng)發(fā)育關(guān)系!痉椒ā繉3亞科共53種灰蝶的線粒體COI和Cytb基因進行序列測定和序列變異分析,同時,基于最大似然法(maximum likelihood,ML)和貝葉斯法(bayesian inference,BI)等建樹方法重建53種灰蝶的系統(tǒng)發(fā)育樹!窘Y(jié)果】串聯(lián)的2個基因共1 431 bp,其中保守位點855個,可變位點576個,簡約信息位點488個;A+T的平均含量為74.5%,明顯高于G+C的平均含量(25.5%)。系統(tǒng)樹顯示,灰蝶亞科以及眼灰蝶亞科均是單系發(fā)生,線灰蝶亞科則為并系群。全部灰蝶物種共分為三大支系:灰蝶亞科為第1支系;眼灰蝶亞科與線灰蝶亞科中的旖灰蝶族(Hypolycaenini)、富麗灰蝶族(Aphnaeini)分別構(gòu)成單系群并互為姊妹群,它們共同構(gòu)成第2支系;線灰蝶亞科中的美灰蝶族(Eumaeini)、玳灰蝶族(Deudorigini)、嬈灰蝶族(Arhopalini)和線灰蝶族(Theclini)構(gòu)成第3支系,其親緣關(guān)系為:(((線灰蝶族+嬈灰蝶族)+玳灰蝶族)+美灰蝶族)!窘Y(jié)論】本研究涉及的3個灰蝶亞科中,灰蝶亞科是一個獨立的支系,眼灰蝶亞科與線灰蝶亞科之間有較近的親緣關(guān)系,但它們內(nèi)部主要類群間的系統(tǒng)發(fā)育關(guān)系還需要進一步的研究。
[Abstract]:[objective] to study the phylogeny of three major subfamilies (Lycaeninaeae, Theclinaeae) and Polyommatinia (Polyommatina), which are closely related to the family Cynidae in China, and to study their phylogeny on the basis of mitochondrial gene sequence data. [methods] Mitochondrial COI and Cytb genes of 53 species in 3 subfamilies were sequenced and sequenced. At the same time, the phylogenetic tree of 53 species of grey butterflies was reconstructed based on maximum likelihood method (MLL) and Bayesian inferenceBI (Bayesian method). [results] two genes in tandem were 1 431 BP, including 855 conserved loci and 576 variable loci. The average content of reduced information sites at 488 sites was 74.5, which was significantly higher than that of GC. The phylogenetic tree showed that both the subfamily and the subfamily Erythroidae were monophylogenetic, while the subfamily of Cynopteridae was a union group. All the species were divided into three branches: the first branch of the subfamily, the second branch of Hypolycaeniniidae, the Aphnaeinii, which formed a monophyletic group, and the second branch of Aphnaeiniae, respectively, and the second branch of Hypolycaeniniidae and Hypolycaeniniidae. Eumaeiniae, Deudoriginius, Arhopalinii and Theclinii from the subfamily Eumaeiniidae form the third branch. The phylogenetic relationship of the three subfamilies in this study was that of the three subfamilies in this study, one is an independent branch, and the other is the family of Dailai, which is a member of the Clariidae. [conclusion] among the three subfamilies in this study, the subfamily is an independent branch. There is a close relationship between the subfamilies of the Orydidae and the subfamilies, but the phylogenetic relationship among the main groups needs further study.
