6株豬流行性腹瀉病毒貴州株S基因序列分析

【摘要】:為了解貴州省豬流行性腹瀉病毒(porcine epidemic diarrhea virus,PEDV)S基因的特點,掌握其遺傳變異情況,本試驗設(shè)計3對特異性引物對6株P(guān)EDV貴州株S基因進(jìn)行全基因RT-PCR擴增、克隆及序列測定;應(yīng)用生物信息學(xué)軟件將6株P(guān)EDV貴州流行株與參考毒株進(jìn)行同源性與系統(tǒng)進(jìn)化分析。結(jié)果顯示,6株P(guān)EDV貴州流行株S基因的全基因長為4 161bp,編碼1 387個氨基酸,與國內(nèi)外PEDV參考毒株核苷酸同源性在93.3%~98.8%之間,氨基酸同源性在91.6%~98.9%之間;進(jìn)化樹分析結(jié)果顯示,6株流行毒株與美國毒株、越南毒株和山東毒株親緣關(guān)系較近;與CV777株、LZC、Brl、83P-5親緣關(guān)系較遠(yuǎn)。結(jié)果表明,PEDV貴州地方流行毒株S基因編碼的氨基酸存在一定程度變化,近年來呈現(xiàn)變異趨勢。本研究可為貴州省PEDV的防控提供一定的理論依據(jù)。
[Abstract]:In order to understand the characteristics and genetic variation of porcine epidemic diarrhea virus (PEDVS) gene of porcine epidemic diarrhea virus in Guizhou Province, three pairs of specific primers were designed to amplify, clone and sequence the S gene of 6 strains of PEDV Guizhou strain. Bioinformatics software was used to analyze the homology and phylogenetic analysis of 6 PEDV epidemic strains and reference strains. The results showed that the total length of S gene was 4 161 BP, encoding 1 387 amino acids. The nucleotide homology of S gene was 93. 398. 8% and amino acid homology was 91.6%. The results of phylogenetic tree analysis showed that 6 epidemic strains were closely related to the American strain, Vietnam strain and Shandong strain, and far related to the CV777 strain LZCCnBrlC83P-5. The results of phylogenetic tree analysis showed that the 6 strains were closely related to the American strain, the Vietnam strain and the Shandong strain. The results showed that the amino acids encoded by S gene of PEDV endemic strain in Guizhou were changed to a certain extent and showed a trend of variation in recent years. This study can provide some theoretical basis for the prevention and control of PEDV in Guizhou province.
