湖羊NR5A1基因SNPs篩選及其與產(chǎn)羔數(shù)的關(guān)聯(lián)分析

【摘要】:為探討NR5A1基因能否作為湖羊產(chǎn)羔性狀的候選基因,同時尋找與湖羊繁殖性能相關(guān)的分子標記,以高繁殖力湖羊為研究對象,采用DNA池結(jié)合測序方法篩選NR5A1基因在湖羊中的SNPs位點,并利用PCR-RFLP和AS-PCR方法進行多態(tài)位點分型,利用SPSS軟件分析不同基因型與湖羊產(chǎn)羔數(shù)的相關(guān)性。結(jié)果表明,在湖羊NR5A1基因中檢測到6個SNPs位點,分析發(fā)現(xiàn)g.6052A/G位點為Taq I酶切位點,其余5個位點處沒有酶切位點存在。在湖羊群體中,g.3362G/C(GG、GC、CC)、g.4342G/A(GG、GA、AA)、g.4666C/T(CC、CT、TT)、g.6052A/G(GG、AG、AA)和g.6991T/G(TT、TG、GG)位點均檢測到3種基因型,而g.5699C/G位點在湖羊群體中僅發(fā)現(xiàn)2種基因型GG和GC。6個位點的多態(tài)信息含量(PIC)從0.306到0.375不等,均屬于中度多態(tài)(0.25PIC0.50),雜合度(He)從0.377到0.500,在這6個SNPs位點中,僅g.6052A/G位點處于Hardy-Weinberg平衡狀態(tài)。連鎖分析發(fā)現(xiàn),g.3362G/C和g.6052A/G 2位點在湖羊群體中緊密連鎖,二者共構(gòu)建了8種單倍型組合,AAGG為主要的單倍型,占總數(shù)的35.90%,AGCG和AACG次之,分別占總數(shù)的20.30%和18.75%。關(guān)聯(lián)分析發(fā)現(xiàn)g.6052A/G位點GG型湖羊的平均產(chǎn)羔數(shù)顯著高于AA型(P0.05)和AG型(P0.05),其余位點的各基因型在湖羊的頭胎產(chǎn)羔數(shù)、二胎產(chǎn)羔數(shù)、三胎產(chǎn)羔數(shù)和平均產(chǎn)羔數(shù)間差異均不顯著。AAGG單倍型和AACG單倍型的二胎產(chǎn)羔數(shù)比AGCG單倍型的二胎產(chǎn)羔數(shù)分別多0.40(P0.05)和0.53(P0.01),差異顯著。說明,NR5A1基因?qū)虍a(chǎn)羔數(shù)有一定的影響,g.6052A/G位點、g.3362 G/C和g.6052A/G連鎖可作為湖羊繁殖性能的有效遺傳標記,用于湖羊的分子選育。
[Abstract]:In order to investigate whether NR5A1 gene can be used as candidate gene for lambing traits of Hu sheep, and to search for molecular markers related to reproduction performance of Hu sheep, the high fecundity Hu sheep was used as the research object. The SNPs loci of NR5A1 gene in Hu sheep were screened by DNA pool and sequencing. The polymorphic loci were classified by PCR-RFLP and AS-PCR. The correlation between different genotypes and litter size of Hu sheep was analyzed by SPSS software. The results showed that six SNPs loci were detected in the NR5A1 gene of Hu sheep. The g.6052A/G loci were found to be Taq I digested sites, while the other 5 loci were not found. In the Hu sheep population, three genotypes were detected, and the polymorphic information contents of the two genotypes (GG and GC.6 loci) were found in the Hu sheep population. The polymorphic information content of the two genotypes (GG and GC.6) ranged from 0.306 to 0.375, and the polymorphic information contents of the GG and GC.6 loci were only found in the Hu sheep population. All of them belong to 0.25PIC0.50, and the heterozygosity is from 0.377 to 0.500.The only g.6052A/G loci in the six SNPs loci are in the Hardy-Weinberg equilibrium state. Linkage analysis showed that the loci of G. 3362G / C and g.6052A/G 2 were closely linked in Hu sheep population. Eight haplotypes were constructed, accounting for 35.90% and 18.75% of the total, respectively. Correlation analysis showed that the average litter size of GG type Hu sheep at g.6052A/G locus was significantly higher than that of AA type P 0.05) and AG type P 0.05%. The other genotypes were in the first litter size and second litter size of Hu sheep. There was no significant difference in the number of lambs and the average number of lambs. The second litter size of the haplotypes of AAGG and AACG haplotypes was 0.40g / P0.05 and 0.53g / 0.01respectively more than that of AGCG haplotypes, and the difference was significant. These results suggest that the linkage of NR5A1 gene to litter size of Hu sheep can be used as an effective genetic marker for breeding performance of Hu sheep and can be used in molecular breeding of Hu sheep. The linkage of G. 6052A / G / G locus and g.6052A/G can be used as an effective genetic marker for breeding performance of Hu sheep.
