天津海河流域水中細(xì)菌抗消毒劑基因檢測與大腸桿菌對苯扎溴銨抗性研究

【摘要】:目的了解天津海河流域水體中細(xì)菌抗消毒劑基因攜帶情況與大腸桿菌對季銨鹽類消毒劑抗性水平,為季銨鹽類消毒劑安全性評價(jià)提供依據(jù)。方法采用PCR技術(shù)和肉湯稀釋法,對海河流域水樣進(jìn)行抗消毒劑基因檢測與大腸桿菌對季銨鹽類抗性水平進(jìn)行研究。結(jié)果從海河流域8處水樣中,均檢出qacE、qacE△1和qacG基因,其中6處水樣檢出ydgE和ydgF基因,4處水樣檢出sugE(p)和emrE基因,所有水樣中均未檢出qacA/B基因。苯扎溴銨對海河水中細(xì)菌的MIC值和MBC值均≥90 mg/L,高于大腸桿菌標(biāo)準(zhǔn)株的60 mg/L。結(jié)論天津海河流域存在抗苯扎溴銨的細(xì)菌,細(xì)菌季銨鹽類消毒劑抗性基因攜帶率較高。
[Abstract]:Objective to investigate the gene carrying of bacterial antidisinfectant and the resistance level of Escherichia coli to quaternary ammonium disinfectants in Haihe River basin of Tianjin, so as to provide the basis for the safety evaluation of quaternary ammonium disinfectants. Methods PCR technique and broth dilution method were used to detect the disinfectant gene and the resistance level of Escherichia coli to quaternary ammonium salt. Results from 8 water samples of Haihe River Basin, qace1 and qacG genes were detected in all water samples, among them, ydgE and ydgF genes were detected in 4 water samples in 6 water samples, and qacA/B genes were not detected in all water samples. The MIC and MBC values of benzalkonium bromide to bacteria in Haihe River water were more than 90 mg / L, which was higher than that of Escherichia coli standard strain (60 mg / L). Conclusion bacteria resistant to benzalkonium bromide exist in Haihe River Basin of Tianjin, and the rate of carrying resistant genes of bacterial quaternary ammonium disinfectants is high.
