青海地區(qū)藏漢民族子癇前期與HLA-G基因非編碼區(qū)多態(tài)性的相關(guān)性研究

【摘要】:目的:通過(guò)研究青海地區(qū)藏漢民族子癇前期患者與HLA-G基因非編碼區(qū)多態(tài)性的相關(guān)性,從而篩選出青海地區(qū)藏漢民族孕婦易感子癇前期所對(duì)應(yīng)的HLA-G多態(tài)性位點(diǎn),為子癇前期的病因?qū)W研究提供依據(jù),指導(dǎo)臨床對(duì)子癇前期的預(yù)防。方法:收集2015年4月到2016年10月期間在青海大學(xué)附屬醫(yī)院產(chǎn)科分娩的藏漢民族正常妊娠孕婦及子癇前期患者的血液標(biāo)本共348例,采用DNA提取、聚合酶聯(lián)反應(yīng)技術(shù)、瓊脂糖凝膠電泳及DNA測(cè)序等方法找出HLA-G基因非編碼區(qū)存在的多態(tài)性位點(diǎn),進(jìn)而通過(guò)相應(yīng)的統(tǒng)計(jì)學(xué)方法分析四組間各位點(diǎn)的等位基因及基因型分布是否存在差異,最后得出相應(yīng)的結(jié)論。結(jié)果:四組間HLA-G的Exon8 14bp插入/缺失多態(tài)性分布差異無(wú)統(tǒng)計(jì)學(xué)意義,而將基因型合并后發(fā)現(xiàn)II基因型和ID/DD基因型分布在漢族正常妊娠組和藏族正常妊娠組之間差異具有統(tǒng)計(jì)學(xué)意義;在HLA-G的5-URR區(qū)域發(fā)現(xiàn)6個(gè)單核苷酸多態(tài)性位點(diǎn),其中2個(gè)(-289、-274)位點(diǎn)多態(tài)性在四組間分布差異具有統(tǒng)計(jì)學(xué)意義,兩兩比較后-289位點(diǎn)的等位基因分布在漢族正常妊娠組和漢族子癇前期組之間有統(tǒng)計(jì)學(xué)差異,-274位點(diǎn)的基因型和等位基因在漢族正常妊娠組和藏族正常妊娠組之間有統(tǒng)計(jì)學(xué)差異,-289/-274位點(diǎn)聯(lián)合基因型在漢族正常妊娠組和漢族子癇前期組之間有統(tǒng)計(jì)學(xué)差異。結(jié)論:HLA-G基因Exon8 14bp插入/缺失位點(diǎn)多態(tài)性和HLA-G基因5-URR的-274位點(diǎn)多態(tài)性可能與民族差異有關(guān),而與青海地區(qū)藏漢族孕婦子癇前期的易感性的關(guān)系還需進(jìn)一步擴(kuò)大樣本數(shù)量來(lái)探討;HLA-G基因5-URR的-289位點(diǎn)多態(tài)性可能與青海地區(qū)漢族子癇前期的易感性有關(guān);HLA-G基因5-URR的-289/-274位點(diǎn)聯(lián)合多態(tài)性可能與青海地區(qū)漢族子癇前期的易感性有關(guān)。
[Abstract]:Objective: to study the association between preeclampsia and non-coding region polymorphism of HLA-G gene in Tibetan and Han nationality preeclampsia patients in Qinghai area, and to screen out the HLA-G polymorphism corresponding to preeclampsia in Tibetan and Han nationality pregnant women in Qinghai area. To provide the basis for the etiological study of preeclampsia and to guide the clinical prevention of preeclampsia. Methods: from April 2015 to October 2016, the blood samples of normal pregnant women and preeclampsia were collected from 348 pregnant women and preeclampsia patients who were delivered by obstetrics in the affiliated Hospital of Qinghai University. DNA extraction and polymerase chain reaction (PCR) were used. Agarose gel electrophoresis and DNA sequencing were used to identify the polymorphic loci in the non-coding region of the HLA-G gene, and then the alleles and genotypes of the four groups were analyzed by statistical methods. Finally, the corresponding conclusions are drawn. Results: there was no significant difference in the distribution of Exon8 14bp insertion / deletion polymorphism in HLA-G among the four groups. It was found that the distribution of genotype II and ID/DD genotype was statistically significant between normal pregnancy group of Han nationality and normal pregnancy group of Tibetan nationality, and six single nucleotide polymorphism loci were found in 5-URR region of HLA-G. There were significant differences in the distribution of two loci polymorphism among the four groups. The allele distribution of -289 locus was significantly different between normal pregnancy group and preeclampsia group of Han nationality. The genotype and allele of -274 locus were found in normal pregnancy group of Han nationality and normal pregnancy group of Tibetan nationality. Statistical difference was found between Han normal pregnancy group and Han preeclampsia group. Conclusion the polymorphism of insertion / deletion locus of Exon8 14bp of HLA-G gene and the polymorphism of 5-URR locus of HLA-G gene may be related to ethnic differences. The relationship between preeclampsia and preeclampsia in Tibetan and Han women in Qinghai area needs to be further expanded to explore the relationship between HLA-G gene 5-URR polymorphism and preeclampsia preeclampsia preeclampsia (5-URR) gene in Qinghai Han nationality. The-289%-274 locus polymorphism may be associated with preeclampsia susceptibility in the Han nationality in Qinghai.
