綿果薺CBF基因啟動子克隆及誘導特性分析

【摘要】:CBF基因不僅是植物CBF抗冷途徑的樞紐,而且可以提高植物的抗冷能力。而植物的抗冷能力的提高是由于CBF基因調控下游抗冷基因的表達來實現(xiàn)的。本研究根據(jù)前期已克隆得到的綿果薺Ll CBF基因(JQ687132)序列,利用TAIL-PCR方法從新疆早春短命植物綿果薺基因組DNA中分離得到Ll CBF基因編碼區(qū)上游1 172 bp啟動子序列;將該序列連入克隆載體并測序,通過生物信息學分析表明,該序列含TATAbox、CAAT-box等典型的真核生物核心啟動子元件,同時具有等逆境響應相關元件。在煙草葉片中的瞬時表達分析,進一步表明,在低溫、干旱及鹽脅迫誘導下該啟動子序列具有驅動GUS報告基因表達的特性。本研究對新疆早春短命植物種質資源的開發(fā)利用及豐富作物逆境基因工程誘導啟動子資源具有重要的實踐意義。
[Abstract]:CBF gene is not only the hinge of plant CBF cold resistance pathway, but also can improve the cold resistance ability of plants. The improvement of plant cold resistance is due to the regulation of downstream cold resistance gene expression by CBF gene. Based on the previously cloned Ll CBF gene JQ687132) sequence, 1 172 BP promoter sequence was obtained from the genomic DNA of Cardamine cardamine, a short-lived plant in early spring in Xinjiang, by TAIL-PCR method. The sequence was inserted into the clone vector and sequenced. Bioinformatics analysis showed that the sequence contained typical eukaryote core promoter elements such as TATAbox and CAAT-box. The transient expression analysis in tobacco leaves further showed that the promoter sequence had the characteristics of driving the expression of GUS reporter gene under the induction of low temperature, drought and salt stress. This study is of great practical significance for the development and utilization of germplasm resources of short-lived plants in early spring in Xinjiang and the enrichment of genetic engineering induced promoter resources for crop stress.
