大口黑鱸肌球蛋白重鏈基因SNPs的篩選及與生長性狀的關(guān)聯(lián)

【摘要】:肌球蛋白是肌肉細(xì)胞的重要組成成分,影響肌纖維的組成和肌肉生長。為了研究肌球蛋白重鏈(MYH)基因多態(tài)性與大口黑鱸生長性狀的相關(guān)性,本研究采用PCR技術(shù)擴(kuò)增得到編碼區(qū)序列全長為9759 bp的大口黑鱸MYH基因,該基因包含37個(gè)外顯子和36個(gè)內(nèi)含子,編碼1940個(gè)氨基酸。采用直接測(cè)序法在MYH基因上篩選到8個(gè)單核苷酸多態(tài)性標(biāo)記(SNP)位點(diǎn)(A-305G、G-558C、A-2784C、A-2816G、T-4765A、C-6206T、C-6811T和G-6935T),有4個(gè)位于外顯子上,其中2個(gè)屬于同義突變。用SNa Pshot方法對(duì)從同批繁殖、同塘養(yǎng)殖的大口黑鱸"優(yōu)鱸1號(hào)"群體中隨機(jī)選取的430尾個(gè)體中各位點(diǎn)的基因型進(jìn)行檢測(cè)。結(jié)果顯示,內(nèi)含子上的A-2784C和A-2816G位點(diǎn)完全連鎖,所有位點(diǎn)在大口黑鱸"優(yōu)鱸1號(hào)"群體中的平均有效等位基因數(shù)、平均觀測(cè)雜合度和平均期望雜合度分別為1.635、0.406和0.373,僅C-6206T、C-6811T和T-4765A位點(diǎn)的基因型頻率分布符合Hardy-Weinberg定律。采用一般線性模型分析各位點(diǎn)與大口黑鱸生長性狀之間的相關(guān)性,研究發(fā)現(xiàn),C-6811T位點(diǎn)CC基因型個(gè)體的體質(zhì)量和全長顯著大于TT基因型,CC基因型個(gè)體的體高和尾柄長顯著大于CT和TT基因型,其余位點(diǎn)不同基因型個(gè)體間的生長性狀均不存在顯著差異。C-6811T位點(diǎn)與生長性狀顯著相關(guān),可作為大口黑鱸分子標(biāo)記輔助育種的候選標(biāo)記。
[Abstract]:Myosin is an important component of muscle cells, which affects muscle fiber composition and muscle growth. In order to study the association between myosin heavy chain myosin (Myh) gene polymorphism and growth traits of large mouth black bass, the MYH gene with coding region of 9759 BP was amplified by PCR technique. The gene contains 37 exons and 36 introns, encoding 1940 amino acids. Eight single nucleotide polymorphisms (SNPs) loci (A-305GN G-558CU A-2816GN T-4765TNC-6811T and G-6935T) were screened on MYH gene by direct sequencing. Four of them were located on exons, 2 of which were synonymous mutations. SNa Pshot method was used to detect the genotypes of each of the 430individuals randomly selected from the population of "you Perch 1", which was bred in the same batch and cultured in Tongtang. The results showed that the A-2784C and A-2816G loci on the intron were completely linked, and the average effective allele number of all the loci in the population of "you Perch 1" was found. The average observed heterozygosity and average expected heterozygosity were 1.6350.406 and 0.373, respectively. Only the genotype frequencies of C-6206TX C-6811T and T-4765A loci were in accordance with Hardy-Weinberg 's law. A general linear model was used to analyze the correlation between the growth traits of Lateolchus japonicus and each point. It was found that the body weight and total length of CC genotype individuals at C-6811T locus were significantly higher than those of CT and TT genotypes, and the body height and tail stalk length of CC genotype individuals with TT genotype were significantly higher than those of CT and TT genotypes. There was no significant difference in growth traits among individuals with different genotypes in other loci. C-6811T loci were significantly correlated with growth traits, and could be used as candidate markers for molecular marker-assisted breeding of large mouth black bass.
