【摘要】：野生二粒小麥(Triticum dicoccoides K錸rn.,AABB,2n=4x=28)NAM-B1基因的表達(dá)可以加速植株衰老,促進(jìn)葉片營(yíng)養(yǎng)向籽粒轉(zhuǎn)移,提高籽粒蛋白質(zhì)、鐵、鋅等物質(zhì)含量。本研究采用同源克隆方法從6份伊斯帕汗小麥(T.ispahanicum Heslot,AABB,2n=4x=28)中克隆得到5個(gè)NAM-B1基因。它們具有典型的NAM基因核苷酸結(jié)構(gòu)特點(diǎn),均含有3個(gè)外顯子和2個(gè)內(nèi)含子。與已報(bào)道的NAM-B1基因比對(duì)分析發(fā)現(xiàn),居群PI330548中的NAM-B1基因因核苷酸序列上第11位點(diǎn)T的插入而引起移碼突變,具有無功能型基因的結(jié)構(gòu)特征,其他4個(gè)則具有功能類型基因的結(jié)構(gòu)特征。4個(gè)功能類型NAM-B1基因的推導(dǎo)氨基酸序列一致性高達(dá)99.7%,它們之間僅有5個(gè)氨基酸的變異。供試的6份伊斯帕汗小麥之間籽粒蛋白質(zhì)含量(GPC)存在顯著差異。與高GPC材料PI346782相比,低GPC材料PI572904的NAM-B1基因編碼氨基酸序列中存在2個(gè)位點(diǎn)的變異,分別為NAC域內(nèi)的C亞區(qū)第88位Q→R和TAR區(qū)域第364位P→R的替換。推測(cè)伊斯帕汗小麥GPC的變異與其NAM-B1基因的這些突變存在一定關(guān)系。
[Abstract]:The expression of NAM-B1 gene in Triticum dicoccoides K rhenium rn. AABB _ 2N ~ (2 +) N ~ (2 +) N ~ (2 +) ~ (28) N ~ (2 +) can accelerate plant senescence, promote leaf nutrient transfer to grain, and increase grain protein, iron, zinc and other substances.In this study, five NAM-B1 genes were cloned from 6 Espa Khan wheat (T. ispahanicum Heslotlota, AABB _ (2) N ~ (4) x) by homologous cloning method.They have typical nucleotide structure of NAM gene and contain 3 exons and 2 introns.Comparing with the reported NAM-B1 gene, it was found that the NAM-B1 gene in the population PI330548 was caused by the insertion of T at site 11 on the nucleotide sequence, resulting in a frameshift mutation, which had the structural characteristics of a non-functional gene.The deduced amino acid sequences of the four functional types of NAM-B1 genes were 99.7 consistent, and there were only 5 amino acid variations between them.There was a significant difference in grain protein content (GPC) among 6 Espa Khan wheat lines.Compared with the high GPC material PI346782, there were two mutations in the amino acid sequence of the NAM-B1 gene of the low GPC material PI572904, which were the replacement of the 88th QR of the C subregion and the 364th PERR of the TAR region in the NAC domain.It is speculated that the variation of GPC in Espa Khan wheat is related to the mutation of its NAM-B1 gene.
