綠頭野鴨蛋品質(zhì)研究及蛋殼強(qiáng)度相關(guān)基因的篩選

【摘要】:本試驗選取體重、周齡一致的100只綠頭野鴨,對其蛋品質(zhì)進(jìn)行測定并進(jìn)行相關(guān)分析,測定指標(biāo)包括蛋殼顏色、蛋殼強(qiáng)度、蛋殼厚度、蛋形指數(shù)、蛋白高度、哈氏單位和蛋黃顏色,同時通過轉(zhuǎn)錄組測序篩選了與蛋殼強(qiáng)度相關(guān)的候選基因。結(jié)果表明:不同蛋殼顏色的鴨蛋之間蛋形指數(shù)、蛋殼強(qiáng)度、蛋殼厚度、蛋白高度和哈氏單位有顯著差異(P0.05);綠頭鴨大部分蛋品質(zhì)性狀之間相關(guān)性顯著,其中蛋殼厚度與蛋殼強(qiáng)度呈顯著正相關(guān),相關(guān)系數(shù)為0.8742,而蛋殼顏色作為蛋殼強(qiáng)度的衡量指標(biāo)時,白殼組強(qiáng)度最大,綠殼組次之,黃綠殼組最小(P0.01);通過轉(zhuǎn)錄組測序?qū)Φ皻?qiáng)度進(jìn)行差異表達(dá)模式對比分析,共找到差異表達(dá)基因325個,其中149個基因表達(dá)量上調(diào),176個基因表達(dá)量下調(diào),與蛋殼強(qiáng)度相關(guān)的基因有CACNA1I、CACNB4、CALN、CACNA1S、LOC101801793等17個基因,這些基因主要參與了鈣離子跨膜轉(zhuǎn)運、鈣離子結(jié)合、鈣離子釋放等生物學(xué)過程,本研究結(jié)果為進(jìn)一步研究蛋殼強(qiáng)度差異的分子機(jī)理提供了基礎(chǔ)。
[Abstract]:The egg quality of 100 mallard ducks of the same weight and age was measured and analyzed, including eggshell color, eggshell strength, eggshell thickness, egg shape index and protein height.At the same time, the candidate genes related to eggshell strength were screened by transcriptome sequencing.The results showed that there were significant differences in egg shape index, egg shell strength, egg shell thickness, protein height and Hardy unit between duck eggs with different eggshell colors, and the correlation between most egg quality traits of mallard ducks was significant.There was a significant positive correlation between eggshell thickness and eggshell strength, the correlation coefficient was 0.8742. When eggshell color was taken as an index of eggshell strength, the intensity of white shell group was the largest, followed by green shell group.A total of 325 differentially expressed genes, including 149 up-regulated genes and 176 down-regulated genes, were found by comparative analysis of differentially expressed patterns of eggshell intensity by transcriptome sequencing.There are 17 genes related to eggshell strength, such as CACNA1I, CACNB4, CACNA1ShlLOC101801793 and so on. These genes are mainly involved in the biological processes of calcium ion translocation, calcium binding, calcium release and so on.The results provide a basis for further study on the molecular mechanism of eggshell strength difference.
