MC1R與Slc7a11基因多態(tài)性與哈薩克羊毛色相關(guān)的研究

【摘要】:本研究采集新疆伊犁哈薩克自治州特克斯縣4種不同被毛顏色2~3周歲的220只哈薩克母羊血樣用于提取DNA,其中黑色被毛羊52只,棕色被毛羊52只,青灰色被毛羊59只,白色被毛羊57只;并采集綿羊左側(cè)肩胛皮膚樣本,每種毛色各10個,用于組織形態(tài)學(xué)觀察。本研究運用PCR-SSCP技術(shù)及測序技術(shù)對MC1R基因及Slc7a11基因多態(tài)性及其對哈薩克羊毛色的影響進(jìn)行研究。運用皮膚樣本制作切片用于不同毛色組織形態(tài)學(xué)觀察。結(jié)果表明:MC1R基因突變位點在白色和棕色被毛群體中,只存在BB基因型;而在黑色和青灰色被毛群體中,存在AA、AB和BB 3種基因型;且AB基因型均為優(yōu)勢基因型,A等位基因為優(yōu)勢等位基因。由組織形態(tài)學(xué)觀察可知,黑色和青灰色被毛組織毛囊分布極為相似,毛囊分布稀疏且皮脂腺發(fā)達(dá);棕色被毛最具特色,毛囊叢分布明顯,皮脂腺分布不發(fā)達(dá);白色被毛毛囊分布最為密集,且皮脂腺分布密集并且最為發(fā)達(dá)。經(jīng)χ~2檢驗,MC1R基因的基因型頻率在4種毛色羊中的分布差異極顯著(P0.01)。Slc7a11基因位點在伊犁哈薩克羊群體中發(fā)現(xiàn)其不存在多態(tài)性。
[Abstract]:In this study, the blood samples of 220 Kazak sheep of 2 or 3 years of age were collected from Tekes County, Yili Kazak Autonomous Prefecture, Xinjiang. Among them, 52 were black wool sheep, 52 brown wool sheep and 59 blue grey wool sheep.The skin samples of left shoulder blades of 57 white wool sheep were collected, 10 of each color were used for histomorphological observation.In this study, PCR-SSCP and sequencing techniques were used to study the polymorphism of MC1R gene and Slc7a11 gene and their effects on the color of Kazak wool.Skin samples were used to make sections for histomorphological observation of different coat colors.The results showed that there were only BB genotypes in white and brown hairy populations, but only three genotypes in black and green grey hairy populations.The AB genotype was the dominant genotype and the allele was the dominant allele.According to histomorphology, the hair follicles of black and gray hair tissues are very similar, the hair follicles are sparse and the sebaceous glands are developed, the brown hair coat is the most characteristic, the hair follicle plexus is obviously distributed, and the sebaceous gland is not developed.White hair follicles are most densely distributed and sebaceous glands are densely distributed and most developed.The genotype frequency of MC1R gene was significantly different among the four kinds of wool sheep by 蠂 ~ 2 test. It was found that there was no polymorphism in Yili Kazak sheep population at the locus of P0.01U. Slc7a11.
