【摘要】：目的探討環(huán)氧化酶(COX)的基因多態(tài)性與阿司匹林抵抗(AR)的關(guān)系。方法選取缺血性腦卒中(IS)患者300例,依據(jù)血小板聚集率分為阿司匹林敏感組(AS組,180例)、阿司匹林半抵抗組(ASR組,84例)和AR組(36例);比較3組一般臨床資料,采用光比濁法檢測(cè)血小板聚集率。應(yīng)用聚合酶鏈反應(yīng)(PCR)和限制性片段長(zhǎng)度多態(tài)性(RFLP)技術(shù)檢測(cè)COX基因多態(tài)性,并比較兩組基因型和等位基因分布差異。結(jié)果 AR組與ASR組COX-1上A-842/C50T有統(tǒng)計(jì)學(xué)差異(P0.05),而COX-2-765GC(rs20417)無(wú)統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論 COX-1上的T等位基因是其突變基因,C/T和TT突變會(huì)增加AR的風(fēng)險(xiǎn),而COX-2-765GC的基因突變與AR無(wú)相關(guān)性。
[Abstract]:Objective to investigate the relationship between the gene polymorphism of cyclooxygenase (COXX) and aspirin resistance to ARA.Methods 300 patients with ischemic stroke (ISI) were divided into three groups according to platelet aggregation rate: aspirin sensitive group (n = 180) and aspirin semi-resistance group (n = 84) and AR group (n = 36).The platelet aggregation rate was measured by phototurbidimetry.Polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) techniques were used to detect the polymorphism of COX gene and to compare the distribution of genotypes and alleles between the two groups.Results there was a significant difference in A-842/C50T on COX-1 between AR group and ASR group (P 0.05), but there was no significant difference in A-842/C50T between AR group and ASR group (P 0.05).Conclusion the T allele in COX-1 is that the mutation of C / T and TT increases the risk of AR, but the mutation of COX-2-765GC has no correlation with AR.
