不同光環(huán)境對(duì)兔視網(wǎng)膜生物鐘基因Cry1、Cry2表達(dá)的影響及中藥干預(yù)作用的研究

【摘要】:目的:本課題通過(guò)動(dòng)物實(shí)驗(yàn)研究,探討不同光環(huán)境對(duì)兔視網(wǎng)膜生物鐘基因Cry1、Cry2表達(dá)的影響。結(jié)合王育良教授提出的日鐘陰陽(yáng)理論,觀(guān)察基于陰陽(yáng)鐘理論上的中藥復(fù)方"日鐘陰陽(yáng)方"對(duì)不同光環(huán)境下兔視網(wǎng)膜鐘基因Cry1、Cry2表達(dá)的干預(yù)作用。方法:72只1月齡雌雄兼?zhèn)涞男挛魈m幼兔,均為清潔動(dòng)物,隨機(jī)標(biāo)號(hào)分為兩大組,分別為4周組和12周組。兩大組又隨機(jī)分為6組,每組6只,即自然光組、正白光LED燈組、全光譜組、自然光中藥組、正白光LED燈中藥組和全光譜中藥組。自然光組:飼養(yǎng)在正常自然光照射的環(huán)境中;正白光LED燈組:飼養(yǎng)在正白光LED燈照射的環(huán)境下;全光譜組;飼養(yǎng)在全光譜燈光環(huán)境中。各中藥組:分別飼養(yǎng)在相應(yīng)的光環(huán)境中,并飼養(yǎng)的同時(shí)分別予以"日鐘陰陽(yáng)方"灌胃,補(bǔ)陽(yáng)方于每日10:00灌入,補(bǔ)陰方于每日22:00灌入。幼兔中藥灌胃劑量參照《藥理實(shí)驗(yàn)學(xué)》中人和動(dòng)物間按體表面積折算的等效劑量比值進(jìn)行換算,按照70kg成人體重標(biāo)準(zhǔn),一副中藥顆粒以300ml水沖調(diào),每日分兩次口服計(jì)算,成人兔(1.5g)每次服藥7ml,幼兔減半為每次3.5ml針管灌胃。自然光組、正白光LED燈組、全光譜組未予以生理鹽水灌胃,正常飼養(yǎng)。測(cè)定實(shí)驗(yàn)光源的光譜參數(shù)。第4周、12周提取相應(yīng)組兔視網(wǎng)膜鐘基因Cry1、Cry2。結(jié)果:4周和12周后,RT-PCR基因定量檢測(cè)基因含量,正白光LED燈組兔視網(wǎng)膜Cry1 mRNA、Cry2 mRNA水平皆明顯降低(P0.01),正白光LED燈中藥組兔視網(wǎng)膜Cry1 mRNA、Cry2 mRNA含量顯著高于正白光LED燈組(P0.01)。而中藥對(duì)自然光組和全光譜組兔視網(wǎng)膜Cry1 mRNA、Cry2 mRNA含量無(wú)影響(P0.05)。免疫印跡法顯示正白光LED燈組兔視網(wǎng)膜Cry1蛋白、Cry2蛋白水平皆明顯降低(P0.01),正白光LED燈中藥組兔視網(wǎng)膜Cry1蛋白、Cry2蛋白含量顯著高于正白光LED燈組(P0.01)。而中藥對(duì)自然光組和全光譜組兔視網(wǎng)膜Cry1蛋白、Cry2蛋白含量無(wú)影響(P0.05)。結(jié)論:正白光LED燈對(duì)視網(wǎng)膜鐘基因Cry1、Cry2表達(dá)水平具有一定的影響。全光譜燈對(duì)鐘基因Cry1、Cry2表達(dá)的影響與自然光相似。"日鐘陰陽(yáng)方"對(duì)正白光LED燈下兔視網(wǎng)膜鐘基因Cry1、Cry2的表達(dá)有一定的干預(yù)作用。
[Abstract]:Aim: to investigate the effects of different light environments on the expression of Cry1 and Cry2 in rabbit retina.Based on the theory of Yin and Yang of Sun Zhong put forward by Professor Wang Yuliang, the effect of Rizhong Yin-yang prescription on the expression of Cry1 and Cry2 in rabbit retina under different light environment was observed.Methods Seventy-two one-month old New Zealand rabbits, both male and female, were randomly divided into two groups: 4-week group and 12-week group.The two groups were randomly divided into 6 groups: natural light group, positive white LED lamp group, full spectrum group, natural light Chinese medicine group, positive white light LED lamp group and full spectrum Chinese medicine group.Natural light group: raised in normal natural light environment; positive white light LED lamp group: raised in positive white light LED lamp environment; full spectrum group; raised in full spectrum light environment.Each traditional Chinese medicine group: feeding in the corresponding light environment, and feeding with "Rizhong Yin and Yang prescription", Buyang prescription at 10:00 daily, nourishing Yin prescription at 22:00 daily.Adult rabbits (1.5 g) were treated with 7 ml each time, and young rabbits were treated with 3.5ml every time.Natural light group, white light LED lamp group and full spectrum group were fed with normal saline.Spectral parameters of experimental light source were measured.The corresponding group of rabbit retina clock gene Cry1 and Cry2 were extracted at the 4th week and 12th week.However, Chinese herbal medicine had no effect on the content of Cry2 mRNA in rabbit retina Cry1 mRNAs in natural light group and full spectrum group.However, Chinese herbal medicine had no effect on the content of Cry1 protein Cry2 in the retina of rabbits in natural light group and full spectrum group.Conclusion: the positive white LED lamp has a certain effect on the expression of Cry1 and Cry2 in retina.The effect of full spectrum lamp on the expression of Cry1 gene Cry2 is similar to that of natural light. "Rizhong Yin-Yang Fang had a certain effect on the expression of Cry1 Cry2 in rabbit retina under positive white LED lamp.
