大豆GmCOL1和GmCOL13啟動(dòng)子和基因組基因的克隆與生物信息學(xué)分析

【摘要】:以大豆天隆1號(hào)為材料提取基因組DNA,分別克隆到長(zhǎng)2 631 bp的Gm COL1啟動(dòng)子和2 809 bp的Gm COL13啟動(dòng)子。此外,還克隆到Gm COL1和Gm COL13基因組基因長(zhǎng)度分別為3 488 bp和2 798 bp,它們分別編碼含348個(gè)和366個(gè)氨基酸的蛋白質(zhì)。利用Plant CARE分析Gm COL1和Gm COL13的啟動(dòng)子序列發(fā)現(xiàn),它們均含有以下調(diào)控元件:生物鐘元件(Circadian)、光響應(yīng)元件(ACEG-box等)、ABA響應(yīng)元件(ABRE)、干旱誘導(dǎo)元件(MBS)、低溫響應(yīng)元件(LTR)。Prot Param分析表明,Gm COL1和Gm COL13蛋白質(zhì)的分子量分別為38.47 k D和40.91 k D,并且都是親水性蛋白。GOR分析顯示,Gm COL1和Gm COL13的二級(jí)結(jié)構(gòu)含有Alpha helix,Extended strand和Random coil。在線網(wǎng)站PROSITE的分析說明,Gm COL1和Gm COL13有兩個(gè)Zinc finger B-box和一個(gè)CCT domain(CO,CO-LIKE and TOC1 domain)結(jié)構(gòu)域。
[Abstract]:Genomic DNA was extracted from soybean Tianlong 1 and cloned into 2 631 BP GM COL1 promoter and 2 809 BP GM COL13 promoter, respectively. The genomic gene length of GM COL1 and GM COL13 were 3 488 BP and 2 798 BP, respectively, which encoded 348 and 366 amino acids, respectively. The promoter sequences of GM COL1 and GM COL13 were analyzed by Plant CARE. They all contain the following regulatory elements: circadian clock element, light response element ACEG-box, ACEG-box, drought induced element, and low temperature response element, LTR.Prot Param analysis shows that the molecular weight of Gm COL1 and GM COL13 protein are 38.47kD and 40.91, respectively.
