POU1F1基因多態(tài)性與藏羊生長性狀的關(guān)聯(lián)性分析

【摘要】:【目的】探討POU1F1基因的多態(tài)性與藏羊生長性狀之間的關(guān)聯(lián)性,尋找與藏羊生長性狀相關(guān)的分子標(biāo)記!痉椒ā坷肈NA測序方法檢測藏羊POU1F1基因的多態(tài)位點并對其進(jìn)行基因分型,分析單一多態(tài)位點不同基因型及雙倍型與藏羊生長性狀的關(guān)聯(lián)性。【結(jié)果】在POU1F1基因第4內(nèi)含子上檢測出1個多態(tài)位點g.14205GA,在第5內(nèi)含子上檢測出2個多態(tài)位點g.15223GA和g.15286AG。關(guān)聯(lián)性分析表明,g.14205GA位點上AA基因型的體質(zhì)量和體長分別極顯著(P0.01)和顯著(P0.05)高于GG和AG基因型;g.15223GA位點上GG基因型的體質(zhì)量顯著高于GA基因型(P0.05),極顯著高于AA基因型(P0.01);g.15286AG位點AA基因型體質(zhì)量極顯著高于GG基因型(P0.01),體高顯著高于GG基因型(P0.05)。單倍型Hap7(-GGA-)的發(fā)生頻率最高,達(dá)到42.40%,其次為單倍型Hap5(-GAA-)和單倍型Hap3(-AGA-),發(fā)生頻率分別為21.50%和14.70%。此外,POU1F1的r2值均小于0.33,說明這3個多態(tài)位點之間不存在強的連鎖性。通過合并基因型發(fā)現(xiàn),雙倍型H7H7(GGA-GGA)的各個生長性狀表現(xiàn)最優(yōu)。【結(jié)論】POU1F1基因的3個SNP位點可用于藏羊生長性狀的選育。
[Abstract]:[objective] to investigate the association between the polymorphism of POU1F1 gene and the growth traits of Tibetan sheep, and to search for the molecular markers related to the growth traits of Tibetan sheep. [methods] the polymorphic loci of POU1F1 gene of Tibetan sheep were detected by DNA sequencing and genotyped. [results] one polymorphic locus (g.14205GA) was detected in intron 4 of POU1F1 gene, two polymorphic loci (g.15223GA) and two loci (g.15223GA) were detected in intron 5 of POU1F1 gene. The correlation analysis showed that the body weight and body length of AA genotype were significantly higher than those of GG and AG genotypes at GG and AG loci, respectively, and the body weight and body length of AA genotypes were significantly higher than those of GA genotypes (P0.05A, P 0.05). The body weight of AA genotype was significantly higher than that of GG genotype P0.01G, and the body height was significantly higher than that of GG genotype (P0.05). Haplotype Hap7G GGA-had the highest frequency of occurrence. The frequency of occurrence was 21.50% and 14.70%, respectively. In addition, the r2 values of POU1F1 were all less than 0.33, indicating that there was no strong linkage among the three polymorphic loci. [conclusion] three SNP loci of POU1F1 gene can be used for breeding growth traits of Tibetan sheep.
