普通煙草CESA基因家族成員的鑒定、亞細(xì)胞定位及表達(dá)分析

【摘要】:纖維素合成酶蛋白(cellulose-synthase proteins,CESA)是一類質(zhì)膜定位蛋白,以蛋白復(fù)合體的形式存在于質(zhì)膜上合成纖維素,在細(xì)胞壁建成和植物生長(zhǎng)發(fā)育過程中起著非常重要的作用。本研究利用CESA蛋白保守域序列PF03552檢索普通煙草(Nicotiana tabacum L.)蛋白序列,并通過擬南芥(Arabidopsis thaliana)10個(gè)CESA蛋白序列在普通煙草基因組數(shù)據(jù)庫(kù)中利用TBLASTN程序進(jìn)行比對(duì),共獲得21條NtCESA基因候選序列,對(duì)這些序列進(jìn)行蛋白序列理化性質(zhì)分析、系統(tǒng)進(jìn)化樹構(gòu)建、基因結(jié)構(gòu)分析、保守結(jié)構(gòu)域及跨膜區(qū)分析和組織表達(dá)模式分析,并對(duì)NtCESA9和NtCESA14兩個(gè)蛋白進(jìn)行了亞細(xì)胞定位實(shí)驗(yàn)。結(jié)果表明:獲得的21條NtCESA蛋白序列的理化性質(zhì)相似;系統(tǒng)進(jìn)化分析將21個(gè)NtCESA基因和10個(gè)At CESA基因分成5個(gè)分支,每一個(gè)分支各成員之間的進(jìn)化相對(duì)保守,基因結(jié)構(gòu)類似,不同分支之間的基因結(jié)構(gòu)差異也較小;NtCESA蛋白結(jié)構(gòu)域相對(duì)保守,都含有CESA蛋白典型的N端鋅指結(jié)構(gòu)、C端跨膜區(qū)和DDD-QXXRW保守功能域;組織表達(dá)分析結(jié)果表明,大部分NtCESA基因在幼苗和成熟期煙草的根、葉、胚芽和愈傷組織中都有表達(dá),同一個(gè)分支中的基因表達(dá)模式基本一致,并且NtCESA基因參與初/次生細(xì)胞壁纖維素的合成與該基因編碼蛋白的跨膜區(qū)數(shù)目存在關(guān)聯(lián),表明NtCESA基因家族成員功能上的復(fù)雜性;亞細(xì)胞定位結(jié)果證實(shí)NtCESA9和NtCESA14為質(zhì)膜定位蛋白。本研究為煙草CESA基因家族功能的深入研究奠定了基礎(chǔ)。
[Abstract]:Cellulose synthase (Cellulose-synthase proteinsCesa) is a kind of plasmalemma localization protein, which exists in the form of protein complex on the plasma membrane to synthesize cellulose. It plays an important role in cell wall building and plant growth and development. In this study, the conserved domain PF03552 of CESA protein was used to search the protein sequence of Nicotiana tabacum L.in common tobacco. Ten CESA sequences of Arabidopsis thaliana were compared by using TBLASTN program in the general tobacco genome database. A total of 21 candidate sequences of NtCESA gene were obtained. The physicochemical properties of these sequences were analyzed and the phylogenetic tree was constructed. Gene structure analysis, conserved domain and transmembrane domain analysis, tissue expression pattern analysis, and subcellular localization of NtCESA9 and NtCESA14 proteins were carried out. The results showed that the obtained 21 NtCESA protein sequences had similar physicochemical properties. Phylogenetic analysis divided 21 NtCESA genes and 10 at CESA genes into five branches. The evolution of each member of each branch was relatively conserved, and the gene structure was similar. The structural differences of NtCESA protein between different branches are also relatively conserved, both of which contain the typical N-terminal zinc finger structure, C-terminal transmembrane region and DDD-QXXRW conserved functional domain, and the results of tissue expression analysis indicate that, Most of the NtCESA genes were expressed in root, leaf, embryo and callus of seedling and mature tobacco, and the same gene expression pattern was found in the same branch. Moreover, NtCESA gene involved in the synthesis of primary / secondary cell wall cellulose was associated with the number of transmembrane regions of the protein encoded by the gene, indicating the functional complexity of the members of the NtCESA gene family. The results of subcellular localization confirmed that NtCESA9 and NtCESA14 were plasma membrane localization proteins, which laid a foundation for the further study of the function of CESA gene family in tobacco.
