甘藍(lán)型油菜DOF蛋白基因的克隆與生物信息學(xué)分析

【摘要】:Dof轉(zhuǎn)錄因子是植物特有的一類轉(zhuǎn)錄因子,含有一個獨特的單鋅指結(jié)構(gòu)域,在植物的生長發(fā)育中起重要的調(diào)控作用。以甘藍(lán)型油菜為試材,采用電子克隆和RT-PCR技術(shù),以期克隆一個具有Zf-Dof結(jié)構(gòu)域的蛋白基因cDNA序列。結(jié)果表明:該基因含有1284bp的開放閱讀框,編碼427個氨基酸,蛋白分子量為46.9kDa,等電點為8.86,為親水性蛋白。經(jīng)序列比對分析表明其與擬南芥CDF3基因有較高同源性,可能具有相似的功能。
[Abstract]:Dof transcription factors are plant specific transcription factors, which contain a unique single zinc finger domain and play an important role in plant growth and development. Using Brassica napus as a test material, electronic cloning and RT-PCR techniques were used. In order to clone the cDNA sequence of a protein gene with Zf-Dof domain, the results showed that the gene contained 1284bp open reading frame, encoding 427amino acids. The protein has a molecular weight of 46.9 kDa and an isoelectric point of 8.86, which is a hydrophilic protein. Sequence alignment analysis shows that the protein has high homology with Arabidopsis CDF3 gene and may have similar function.
