【摘要】:抗菌肽是機體先天免疫的重要組成成分,是許多生物體抵抗外來致病菌侵襲的第1道屏障。Indolicidin是一個13氨基酸殘基肽,從牛的嗜中性粒細胞的胞漿小顆粒分離而來,具有廣譜抗菌性,Rev4是其人工設(shè)計的反向序列。利用經(jīng)典葉盤法,以根癌農(nóng)桿菌A.GV3850介導(dǎo)紫花苜蓿遺傳轉(zhuǎn)化,GUS瞬時表達檢測轉(zhuǎn)化體系建立成功;壓力篩選后葉片基因組DNA的PCR成功檢測到抗菌肽基因條帶,PCR產(chǎn)物回收測序得到13株轉(zhuǎn)化紫花苜蓿。
[Abstract]:Antimicrobial peptide is an important component of innate immunity. It is the first barrier for many organisms to resist the invasion of foreign pathogens. Indolicidin is a 13-amino acid peptide isolated from the cytoplasmic granules of bovine neutrophils. The reverse sequence with broad spectrum antimicrobial activity was established successfully by Agrobacterium tumefaciens A. GV3850 mediated genetic transformation of alfalfa. After pressure screening, 13 transformed alfalfa strains were successfully detected by PCR from genomic DNA of leaves, and 13 strains of alfalfa were recovered and sequenced.
