小麥銹菌碳水化合物酶類基因家族的鑒定與進(jìn)化分析

【摘要】:本研究利用已測(cè)序的條形柄銹菌Puccinia striiformis f.sp.tritici基因組序列信息,結(jié)合已公布的隱匿柄銹菌Puccinia recondita f.sp.tritici和禾柄銹菌Puccinia graminis f.sp.tritici全基因組編碼蛋白序列,運(yùn)用生物信息數(shù)據(jù)庫(kù)和分析軟件對(duì)其碳水化合物酶類(CAZymes)基因家族進(jìn)行了注釋和比較分析。結(jié)果表明,3種小麥銹菌共包含1 232個(gè)CAZymes編碼基因(含有1 279個(gè)模塊)。其中,條形柄銹菌具有明顯較大的CAZymes家族,3種銹菌的CAZymes具有136個(gè)同源基因;相對(duì)于隱匿柄銹菌和禾柄銹菌,條形柄銹菌的GH、GT和AA家族發(fā)生顯著擴(kuò)增,而3種銹菌的PL家族并無(wú)差異;CBM家族則表現(xiàn)為由禾柄銹菌、隱匿柄銹菌到條形柄銹菌的遞增式擴(kuò)增。通過(guò)CAZymes、細(xì)胞壁降解酶、分泌蛋白、PHI、保守基序等不同形式注釋的比較分析表明,對(duì)該基因家族而言,條形柄銹菌相對(duì)于隱匿柄銹菌和禾柄銹菌呈擴(kuò)增現(xiàn)象,隱匿柄銹菌和禾柄銹菌的家族縮減水平存在差異。小麥銹菌CAZymes家族整體呈現(xiàn)的不同形式擴(kuò)增或縮減現(xiàn)象,可能反映了銹菌與寄主長(zhǎng)期協(xié)同進(jìn)化過(guò)程中的寄生適應(yīng)性需求,進(jìn)而反映了其致病性演化機(jī)理。本研究結(jié)果有助于進(jìn)一步全面認(rèn)識(shí)小麥銹菌CAZymes基因家族進(jìn)化在銹菌致病性變異中的作用,為揭示銹菌致病分子機(jī)制提供理論基礎(chǔ)。
[Abstract]:In this study, the genomic sequence information of Puccinia striiformis f.sp.tritici, which has been sequenced, was combined with the published genome coding protein sequences of Puccinia recondita f.sp.tritici and Puccinia graminis f.sp.tritici. Bioinformatics database and analysis software were used to annotate and compare the carbohydrate enzyme CAZymes gene family. The results showed that the three wheat rust species contained 1 232 CAZymes coding genes (including 1 279 modules). There were 136 homologous genes in CAZymes of 3 species of rust species of CAZymes family, compared with that of Puccinia occult and Puccinia graminis, the GHGGT and AA families of Puccinia stripeformis were significantly expanded. But the PL family of the three species of rust showed no difference, but the CBM family showed the incremental amplification from Puccinia graminearum, Puccinia occult to Puccinia striatum, and the cell-wall degradation enzyme was obtained by CAZymes. The comparative analysis of the secretory protein PHIand conserved motifs showed that, for the gene family, Puccinia striatum was more abundant than Puccinia occult and Puccinia graminearum. There were differences in the family reduction level between Puccinia occult and Puccinia graminis. The different forms of expansion or reduction in the CAZymes family of wheat rust might reflect the parasitic adaptability during the long-term coevolution of rust and host. The results of this study are helpful to further understand the role of CAZymes gene family evolution in the pathogenicity variation of wheat rust, and provide a theoretical basis for revealing the molecular mechanism of rust pathogen pathogenicity.
