尼羅羅非魚CD80基因的分子克隆與mRNA表達(dá)分析

【摘要】:CD80是T細(xì)胞活化和增殖過程中不可缺少的膜抗原,為T細(xì)胞的增殖分化提供必要的共刺激信號(hào),在特異性細(xì)胞免疫過程中具有重要作用。本研究從羅非魚脾臟組織中克隆獲得CD80基因的編碼區(qū)序列(Gen Bank登錄號(hào):KU884324;命名為On-CD80),該基因全長810 bp,可編碼269個(gè)氨基酸,理論分子量為29.83 k D,等電點(diǎn)為5.97。氨基酸序列分析顯示On-CD80具有2個(gè)保守的免疫球蛋白樣結(jié)構(gòu)域(Ig-like domain),5'端有一個(gè)典型的信號(hào)肽序列,3'端具有1個(gè)跨膜結(jié)構(gòu)域。系統(tǒng)進(jìn)化樹中羅非魚與伯氏樸麗魚CD80蛋白聚為一支,顯示了較高的同源性。熒光定量PCR分析顯示,On-CD80在健康尼羅羅非魚組織中均有表達(dá),且在脾臟組織中表達(dá)量最高。經(jīng)滅活的無乳鏈球菌免疫后,On-CD80在腎臟、脾臟、胸腺、腸道、鰓、腦組織中的表達(dá)量均顯著上調(diào),且表現(xiàn)出較為明顯的時(shí)間依賴性表達(dá)模式,腸道、鰓和腦分別在8 h、24 h出現(xiàn)表達(dá)高峰,并于48 h、96 h再次出現(xiàn)表達(dá)高峰。脾臟和腎臟、胸腺的On-CD80表達(dá)則分別是在48 h、72 h達(dá)到表達(dá)高峰。綜上所述,On-CD80表達(dá)能被無乳鏈球菌所誘導(dǎo),提示該分子可能參與羅非魚的抗菌免疫應(yīng)答,該結(jié)果為進(jìn)一步研究On-CD80在羅非魚感染無乳鏈球菌過程中的作用奠定了一定的基礎(chǔ)。
[Abstract]:CD80 is an indispensable membrane antigen in the process of T cell activation and proliferation, which provides necessary costimulatory signals for T cell proliferation and differentiation. In this study, the coding region sequence of CD80 gene was cloned from spleen tissue of tilapia, Genin Bank accession number: KU884324, named On-CD80, which is 810 BP in length and encodes 269 amino acids. The theoretical molecular weight is 29.83 KD and the isoelectric point is 5.97.The amino acid sequence analysis shows that On-CD80 has two conserved immunoglobulin-like domains (Ig-like domain) and a typical signal peptide sequence (3'terminal) with a transmembrane domain. The phylogenetic analysis shows that On-CD80 has two conserved immunoglobulin-like domains. The CD80 protein of Tilapia and Puccinia berryi are clustered into one branch in the tree. Fluorescence quantitative PCR analysis showed that On-CD80 was expressed in the tissues of healthy Nile tilapia and had the highest expression in spleen. On-CD80 was detected in kidney, spleen, thymus, intestine and Gill after inactivated streptococcus acus immunization. The expression levels in brain tissues were significantly up-regulated and showed a time-dependent expression pattern. The expression peaks in intestine, Gill and brain were observed at 8 h, 24 h, and 48 h, 96 h, respectively, and in spleen and kidney. The expression of On-CD80 in thymus reached its peak at 48 h and 72 h, respectively. In conclusion, the expression of On-CD80 could be induced by Streptococcus acuminata, suggesting that this molecule may be involved in the antimicrobial immune response of tilapia. The results laid a foundation for the further study of the role of On-CD80 in the process of infection of Streptococcus actinoides in tilapia.
