【摘要】：目的基于全基因組關(guān)聯(lián)分析(GWAS)的研究結(jié)果,初步建立一個2型糖尿病易感基因的局部基因網(wǎng)絡(luò)圖,為今后的基因-基因交互作用,基因-環(huán)境交互作用的研究提供參考。方法該研究的65個候選基因選自經(jīng)過GWAS研究獲得統(tǒng)計學(xué)關(guān)聯(lián)的2型糖尿病易感基因,從中選取具有2個或以上通路的目標(biāo)基因27個,并通過通路的篩選、中間基因的選取,構(gòu)建局部基因網(wǎng)絡(luò),并利用Ai CS6軟件繪制網(wǎng)絡(luò)圖。網(wǎng)絡(luò)構(gòu)建由以下步驟構(gòu)成:(1)通過KEGG、Genecards數(shù)據(jù)庫獲取基因和通路的基本信息;(2)建立初級基因網(wǎng)絡(luò);(3)選取中間基因;(4)構(gòu)建基因二級連接;(5)構(gòu)建局部基因網(wǎng)絡(luò)并繪制網(wǎng)絡(luò)圖。結(jié)果獲得了1個包括27個易感基因和10個中間基因以及一系列連接基因和中間分子的局部基因網(wǎng)絡(luò)圖,該網(wǎng)絡(luò)包含20條生物通路。結(jié)論該研究所得的局部基因網(wǎng)絡(luò)圖較為直觀和綜合地展示β細(xì)胞功能減退和胰島素抵抗易感基因之間的相互關(guān)系,可對深入研究2型糖尿病分子病因機(jī)制提供參考。
[Abstract]:Objective to establish a local gene network map of susceptibility genes of type 2 diabetes mellitus based on the results of GWASs. Methods Sixty-five candidate genes in this study were selected from a statistically associated type 2 diabetes susceptibility gene obtained from the GWAS study, and 27 target genes with 2 or more pathways were selected from the study. And through the selection of the pathway and the selection of the intermediate gene, the local gene network is constructed. Using Ai CS6 software to draw the network map. The network construction consists of the following steps: 1) obtaining basic information of genes and pathways through the KEGGG gene bases. (2) setting up a primary gene network / 3) selecting the intermediate gene 4) constructing a secondary gene link (5). A local gene network consisting of 27 susceptible genes and 10 intermediate genes and a series of linked genes and intermediate molecules were obtained. The network consists of 20 biological pathways. Conclusion the local gene network obtained by this study shows the relationship between 尾 -cell dysfunction and insulin resistance susceptibility genes directly and synthetically. It can provide reference for further study on molecular etiology of type 2 diabetes.
