【摘要】：目的:檢測(cè)牙齦卟啉單胞菌(Porphyromonas gingivalis,P.gingivalis)唾液酸酶基因(PG0352)缺失是否影響其感染上皮細(xì)胞后對(duì)上皮細(xì)胞炎癥相關(guān)小RNA的調(diào)控。方法:厭氧培養(yǎng)P.gingivalis W83及其唾液酸酶基因突變株(△PG0352),以MOI為100∶1的比例分別與永生化牙齦上皮細(xì)胞epi4進(jìn)行共培養(yǎng),PCR array的方法檢測(cè)88個(gè)與炎癥相關(guān)的小RNA的表達(dá)。結(jié)果:△PG0352組epi4的hsa-miR-9、hsa-miR-23a、hsa-miR-23b和hsa-miR-410的表達(dá)分別是P.gingivalis W83組的4.3387、3.3978、4.7930和4.3137倍。結(jié)論:與P.gingivalis W83野生株相比,唾液酸酶基因缺陷株對(duì)牙齦上皮細(xì)胞炎癥相關(guān)小RNA的表達(dá)有不同的調(diào)控作用。
[Abstract]:Objective: to determine whether the deletion of porphyromonas gingivalis-P.gingivalis (PG0352) affects the regulation of small RNA associated with inflammation in epithelial cells after infection with porphyromonas gingivalis W83 and its sialidase gene mutant. Methods: anaerobic culture of P. gingivalis W83 and its sialidase gene mutant. (PG0352). The expression of HSA-miR-9, hsa-miR-23aanhsa-miR-23b and hsa-miR-410 in PG0352 group was detected by co-culture array with epi4 of immortalized gingival epithelial cells. Results: the expression of epi4 in PG0352 group was higher than that in P.gingivalis W83 group. Conclusion: compared with P. gingivalis W83 wild strain, 4.33870.4.7930 and 4.3137.Conclusion: compared with P.gingivalis W83 wild strain, Sialidase gene deficient strain has different regulation on the expression of small RNA associated with inflammation in gingival epithelial cells.
