西瓜噬酸菌Aac-5AAA ATPases基因moxR和ruvB功能研究

【摘要】:AAA ATPases(ATPase associated with diverse cellular activities)是一種與細胞活性相關的酶,廣泛存在于所有生物中,同時具有蛋白酶和分子伴侶活性。為了研究AAA ATPases相關基因缺失對西瓜噬酸菌(Acidovorax citrulli)的影響,本研究通過同源重組雙交換的方法構建西瓜噬酸菌moxR基因以及ruv B基因全缺失突變菌株及互補菌株,測定了突變菌株的致病性、煙草過敏反應、運動性、生長能力、生物膜形成等;并利用實時熒光定量PCR方法測定了AAA ATPases與Ⅲ型分泌系統(tǒng)相關基因(hrc N和hrp E)、毒性蛋白相關基因(trb C和vir B)、鞭毛相關基因(fliR和fli M)、菌毛相關基因(pin C-1和pin C-2)以及AAA ATPases亞家族Ruv B相關基因(ruv A和ruv C)的關系。結果表明,moxR基因以及ruv B基因全缺失突變菌株的致病力降低、運動性減弱、生長能力下降、生物膜形成能力增強、煙草過敏性反應減弱、突變菌株中重要的致病基因hrc N和hrp E表達量顯著下調(diào),表明AAA ATPases相關基因moxR、ruv B在西瓜噬酸菌的致病力上發(fā)揮重要作用。
[Abstract]:AAA ATPases(ATPase associated with diverse cellular actives is a cell-activity related enzyme that is widely present in all organisms and has protease and molecular chaperone activity. In order to study the effect of AAA ATPases related gene deletion on Acidovorax citrullii, In this study, moxR gene and ruv B gene deletion mutant strains and complementary strains were constructed by homologous recombination and double exchange method.
