注意缺陷多動(dòng)障礙轉(zhuǎn)換功能與NRXN1基因的關(guān)聯(lián)研究

【摘要】:目的:探討注意缺陷多動(dòng)障礙(ADHD)轉(zhuǎn)換功能與NRXN1基因的關(guān)聯(lián)。方法:順序納入符合美國(guó)精神障礙診斷與統(tǒng)計(jì)手冊(cè)第4版(DSM-IV)ADHD診斷標(biāo)準(zhǔn)的ADHD兒童756例和年齡匹配的正常對(duì)照兒童133例。采用TMT連線測(cè)試(Trail Making Test)記錄被試者轉(zhuǎn)換功能各指標(biāo),包括數(shù)字連線時(shí)間、數(shù)字連線錯(cuò)誤次數(shù)、數(shù)字字母連線時(shí)間和數(shù)字字母連線錯(cuò)誤次數(shù)。選擇NRXN1基因的兩個(gè)單核苷酸多態(tài)性(rs1592728和rs4971652)位點(diǎn),利用Sequenom基因分型平臺(tái)進(jìn)行基因型檢測(cè)。在全部ADHD病例兒童和正常對(duì)照采用多重線性回歸分析基因型對(duì)ADHD轉(zhuǎn)換功能的影響因素,其次,分別在ADHD組和對(duì)照組中采用多重線性回歸分層分析轉(zhuǎn)換功能與基因型之間的關(guān)聯(lián)。結(jié)果:多重線性回歸顯示轉(zhuǎn)換功能與年齡(β=0.42,P0.001)、智商(β=0.34,P0.001)、罹患ADHD(β=0.08,P=0.004)和rs4971652GG基因型(β=0.06,P=0.039)負(fù)向關(guān)聯(lián);ADHD組轉(zhuǎn)換功能與年齡(β=0.46,P0.001)、智商(β=0.32,P0.001)、rs4971652的GG基因型(β=0.07,P=0.018)負(fù)向關(guān)聯(lián),與ADHD亞型(β=0.06,P=0.033)正向關(guān)聯(lián),ADHD-I型比ADHD-C型患兒受損嚴(yán)重;對(duì)照組轉(zhuǎn)換功能與年齡(β=0.25,P=0.002)、智商(β=0.40,P0.001)負(fù)向關(guān)聯(lián)。結(jié)論:ADHD患兒轉(zhuǎn)換功能可能與NRXN1基因SNPs存在關(guān)聯(lián),GG基因型患者轉(zhuǎn)換功能受損相對(duì)輕。
[Abstract]:Objective: to investigate the association between ADHD conversion function and NRXN1 gene in attention deficit hyperactivity disorder (ADHD). Methods: 756 children with ADHD and normal age-matched children who met the diagnostic criteria of DSM-IVD ADHD in the fourth edition of the American Handbook for the diagnosis and Statistics of Mental Disorder were included in the sequence. In the control group, 133 children were tested with TMT Making test to record each index of the subjects' conversion function. Including digital connection time, number of digital connection errors, digital alphanumeric connection time and number of alphanumeric connection errors. Two single nucleotide polymorphisms of the NRXN1 gene, rs1592728 and rs4971652) were selected. Genotypes were detected by Sequenom genotyping platform. Multiplex linear regression analysis was used to analyze the influence factors of genotype on ADHD conversion function in all children with ADHD and normal controls. Multiple linear regression analysis was used to analyze the correlation between conversion function and genotype in ADHD group and control group respectively. Results: multiple linear regression analysis showed that conversion function and age (尾 -0.42), intelligence quotient (尾 = 0.34), ADHD (尾 -0.08) and rs4971652GG genotype (尾 0.06P ~ (0.039)) were negatively correlated with age (尾 = 0.42), IQ (尾 = 0.34), and rs4971652GG genotype (尾 0.06 P ~ (0.039)). The conversion function of ADHD group was negatively correlated with the GG genotype of age (尾 -0.46, P 0.001), IQ (尾 -0.32, P 0.001, rs4971652) (尾 -0. 07, P 0. 018, P 0. 018), and a negative correlation between the GG genotype and age (尾 -0.36, P 0. 001, P 0. 001). The positive association of ADHD-I with ADHD subtype was more serious than that of ADHD-C, and the transition function of control group was negatively correlated with age (尾 -0.25) and intelligence quotient (尾 _ (0.40) P 0.001). Conclusion there may be a correlation between the conversion function of children with ADHD and SNPs of NRXN1 gene, and the impairment of conversion function in patients with GG genotype is relatively slight.
