牦牛TMEM-18基因多態(tài)性及其與生產(chǎn)性能關(guān)聯(lián)分析

【摘要】:跨膜蛋白18基因(TMEM-18)是一個(gè)末端為低聚嘧啶的基因,與動(dòng)物生長(zhǎng)發(fā)育密切相關(guān)。為了研究牦牛TMEM-18基因多態(tài)性與生產(chǎn)性能的關(guān)系。以192頭天祝雌性牦牛血樣NDA構(gòu)建混合池,擴(kuò)增TMEM-18基因內(nèi)含子1和外顯子5的序列。運(yùn)用BLAST和Chromas軟件分析突變位點(diǎn),運(yùn)用高分辨率熔解曲線分析技術(shù)(HRM)統(tǒng)計(jì)基因型分型,采用SHEsis軟件進(jìn)行基因型頻率和等位基因頻率計(jì)算,同時(shí)對(duì)多態(tài)位點(diǎn)進(jìn)行配對(duì)連鎖不平衡和單倍型分析,運(yùn)用SPSS21.0分析基因多態(tài)位點(diǎn)與生產(chǎn)性能關(guān)聯(lián)。結(jié)果表明,牦牛TMEM-18基因內(nèi)含子1處存在2個(gè)多態(tài)位點(diǎn),分別是861(A/G)和1267(C/T),外顯子5處存在1個(gè)多態(tài)位點(diǎn)為4 447(C/T)。關(guān)聯(lián)分析表明,牦牛TMEM-18基因861(A/G)位點(diǎn)的不同基因型與牦牛的體高、體重、胴體重和屠宰率差異性顯著(P0.05);1267(C/T)位點(diǎn)的不同基因型與牦牛的體高、體重和屠宰率差異性顯著(P0.05),而牦牛TMEM-18基因4447(C/T)位點(diǎn)的不同基因型與牦牛的體斜長(zhǎng)、管圍、胸圍、體重、胴體重、凈肉重、凈肉率和屠宰率均存在顯著性差異(P0.05)。通過(guò)單倍型分析發(fā)現(xiàn)群體中存在3種單倍型組合,即ACC單倍型、GTC單倍型和GTT單倍型,其中ACC單倍型組合個(gè)體數(shù)目明顯多于其他單倍型組合,為優(yōu)勢(shì)單倍型。本實(shí)驗(yàn)揭示TMEM-18基因可作為牦牛生產(chǎn)性能開(kāi)發(fā)的候選分子標(biāo)記,為牦牛遺傳資源的利用、開(kāi)發(fā)與新品種的選育提供依據(jù)。
[Abstract]:Transmembrane protein 18 gene (TMEM-18) is a gene with terminal oligopyrimidine. In order to study the relationship between TMEM-18 gene polymorphism and production performance, the mixed pool was constructed from 192 female yak blood samples with NDA. The sequences of intron 1 and exon 5 of TMEM-18 gene were amplified and the mutation sites were analyzed by BLAST and Chromas software. High resolution melting curve analysis technique was used to calculate genotypes, and SHEsis software was used to calculate the frequency of genotypes and alleles. At the same time, the linkage disequilibrium and haplotype of the polymorphic loci were analyzed, and the correlation between the polymorphic loci and the production performance was analyzed by SPSS21.0. There are two polymorphic loci at intron 1 of TMEM-18 gene in yak, which are 861 A / G) and 1267 C / T respectively. One polymorphic locus was found at exon 5. The correlation analysis showed that the different genotypes of the 861 TMEM-18 gene and the body height and body weight of the yak. The difference of carcass weight and slaughter rate was significant (P0.05). Different genotypes of 1267 C / T locus were significantly different from yak body height, body weight and slaughter rate (P0.05). However, different genotypes of TMEM-18 gene 4447 C / T locus were associated with the body length, tube circumference, chest circumference, body weight, carcass weight and net meat weight of yak. There were significant differences in net meat rate and slaughter rate. Haplotype analysis showed that there were three haplotypes in the population, that is, ACC haplotype GTC haplotype and GTT haplotype. The number of individuals of ACC haplotype combinations was significantly more than that of other haplotypes, which was the dominant haplotype. This study revealed that TMEM-18 gene can be used as candidate molecular markers for the development of yak production performance. To provide the basis for utilization, development and breeding of new varieties of yak genetic resources.
