云南地區(qū)武定雞白血病病毒受體TVB基因型的檢測

【摘要】:通過了解武定雞群白血病病毒受體TVB基因型分布情況,為下一步研究ALV-B、D、E亞群禽白血病抗病育種奠定基礎(chǔ)。試驗以云南地區(qū)三個不同養(yǎng)雞場的168日齡武定雞為研究對象,隨機(jī)分成三個平行組,每組200只。采用PCR-RFLP的方法對TVB受體基因進(jìn)行檢測。結(jié)果:在武定雞群中TVB*S1/S1基因型頻率較高,頻率為0.28~0.355;TVB*RR基因型頻率較低,頻率為0.03~0.065;而其他基因型頻率中等,頻率為0.120~0.205。在各組中TVB*S1等位基因頻率最高,為0.563;最低為TVB*R等位基因,頻率為0.115;TVB*S3頻率介于TVB*S1和TVB*R兩者之間。在這3組內(nèi)TVB*S1/R、TVB*S3/R和TVB*S3/S3基因型之間頻率無顯著性差異(P0.05),TVB*S1/S3、TVB*S1/S1和TVB*R/R基因型頻率低于前面三種基因型,高于TVB*R/R基因型。TVB*S1/S1基因型頻率在組內(nèi)最高,顯著高于其他五種基因型(P0.05),TVB*S1/S3基因型次之。結(jié)果表明:在武定雞群中易感B、D、E亞型禽白血病病毒的基因頻率較高,而抗B、D、E亞群禽白血病毒的基因頻率在中等水平,為后續(xù)抗B、D、E亞群禽白血病病毒選育工作提供遺傳背景資料。
[Abstract]:To investigate the distribution of TVB genotypes of Leukemia virus receptor (TVB) in Wuding chickens, and to study ALV-BUD for the next step. The 168-day-old Wuding chickens from three different chicken farms in Yunnan Province were randomly divided into three parallel groups. The TVB receptor gene was detected by PCR-RFLP. Results: the frequency of TVB*S1/S1 genotype was higher in Wuding chickens. The frequency is 0.28 ~ 0.355; The frequency of TVB*RR genotype was low, 0.03 ~ 0.065; The frequency of other genotypes was moderate (0.120) 0.205. The frequency of TVB*S1 allele was the highest (0.563) in each group. The lowest allele was TVB*R, the frequency was 0.115; The frequency of TVB*S3 was between TVB*S1 and TVB*R. TVB*S1/R was found in these three groups. There was no significant difference in frequency between TVB*S3/R and TVB*S3/S3 genotypes. The frequency of TVB*S1/S1 and TVB*R/R genotypes was lower than that of the former three genotypes, and higher than that of TVB*R/R genotypes. It was significantly higher than that of the other five genotypes (P0.05TV-S1 / S3). The results showed that the gene frequency of the susceptible avian leukemia virus was higher in Wuding chickens than that in the other five genotypes. However, the gene frequency of avian leukemia virus resistant to subgroup BGDU E was at a moderate level, which provided genetic background information for the subsequent selection of avian leukemia virus resistant to subgroup BGDU E.
