利用轉(zhuǎn)錄組測序篩選雞蛋褐殼性狀相關(guān)基因

【摘要】:褐殼雞蛋在許多國家深受消費者喜愛,消費者通常把蛋殼褐色深淺作為評價雞蛋品質(zhì)的重要指標。褐殼雞蛋蛋殼顏色形成受多基因共同調(diào)控,但是具體候選基因及調(diào)控機理尚未明確。因此,本研究以純系褐殼蛋雞為實驗材料,篩選調(diào)控褐殼雞蛋顏色深淺的候選基因。采用轉(zhuǎn)錄組測序技術(shù)對產(chǎn)深褐殼雞蛋和淺褐殼雞蛋的母雞蛋殼腺組織進行分析,結(jié)果顯示,共計有8461個基因在蛋殼腺組織表達,其中34個基因在兩組之間差異表達。功能分析發(fā)現(xiàn),卵轉(zhuǎn)鐵蛋白(ovotransferrin,TF)基因、熱休克蛋白70(heat shock protein,HSP70)基因以及氧化磷酸化通路均與原卟啉Ⅸ合成通路相關(guān),可能影響褐殼雞蛋蛋殼色素原卟啉Ⅸ的合成和積累。
[Abstract]:Brown shell eggs are very popular among consumers in many countries. Consumers usually regard the brown depth of eggs as an important index to evaluate the quality of eggs. The color formation of brown shell eggs is regulated by multiple genes. However, the specific candidate genes and regulatory mechanisms have not been clarified. Therefore, the pure line brown shell layer was used as the experimental material in this study. Screening candidate genes to regulate the color of brown shell eggs. The results showed that the female eggs with deep brown shell eggs and light brown shell eggs were analyzed by transcriptome sequencing. A total of 8461 genes were expressed in eggshell glandular tissues, 34 of which were differentially expressed between the two groups. Heat shock protein 70 (HSP70) gene and oxidative phosphorylation pathway are related to protoporphyrin IX synthesis pathway. It may affect the synthesis and accumulation of pigmented protoporphyrin IX in egg shell of brown shell.
