在患CyHV-2病的異育銀鯽腸道黏膜中膽固醇、膽汁酸代謝通路基因的差異表達(dá)

【摘要】:為了探討CyHV-2疾病條件下異育銀鯽膽汁酸腸肝循環(huán)代謝途徑主要基因表達(dá)活性的變化,以患CyHV-2病的、正常的異育銀鯽腸道黏膜為材料,提取總RNA,采用RNA-Seq測(cè)序、對(duì)單一基因進(jìn)行注釋,并進(jìn)行KEGG富集分析、單一基因差異表達(dá)分析。結(jié)果顯示,腸道黏膜組織有7770個(gè)基因顯著性差異表達(dá),其中,差異表達(dá)上調(diào)基因數(shù)為3335個(gè)、差異表達(dá)下調(diào)的基因數(shù)為4435個(gè),表明CyHV-2病的發(fā)生對(duì)腸道黏膜組織基因表達(dá)產(chǎn)生了重大的影響。病魚膽固醇、膽汁酸生物合成代謝途徑的酶、蛋白質(zhì)的基因顯著性差異表達(dá),顯示腸道黏膜膽固醇、膽汁酸合成代謝受到顯著性影響。參與膽固醇、膽汁酸合成代謝調(diào)節(jié)作用,以及膽固醇、膽汁酸分泌、吸收、轉(zhuǎn)運(yùn)等生理過程的蛋白質(zhì)、酶的基因也是差異表達(dá)下調(diào),膽汁酸腸肝循環(huán)有出現(xiàn)代謝障礙的趨勢(shì)。結(jié)果表明,CyHV-2病的發(fā)生對(duì)腸道黏膜組織基因表達(dá)產(chǎn)生了嚴(yán)重影響,對(duì)膽固醇、膽汁酸的合成代謝途徑、膽汁酸腸肝循環(huán)途徑的基因表達(dá)產(chǎn)生了嚴(yán)重影響,將導(dǎo)致病魚體內(nèi)膽固醇、膽汁酸量的不足;糃yHV-2病病魚血清膽汁酸含量下降了99%、膽固醇含量下降了10%,證實(shí)了上述基因差異表達(dá)的趨勢(shì)。
[Abstract]:In order to investigate the changes of the main gene expression activity of intestinal circulatory metabolism pathway of bile acid in silver crucian carp under the condition of CyHV-2 disease, the intestinal mucosa of normal allogeneic crucian carp with CyHV-2 disease was used as the material. Total RNAs were extracted, RNA-Seq sequencing was used to annotate the single gene, KEGG enrichment analysis was carried out, and the differential expression analysis of single gene was carried out. There were 7770 genes differentially expressed in intestinal mucosa, of which 3335 genes were up-regulated and 4435 genes were down-regulated. The results showed that the occurrence of CyHV-2 disease had a significant effect on the gene expression of intestinal mucosal tissue. The genes of cholesterol, bile acid biosynthesis pathway and protein were significantly differentially expressed in the diseased fish. The results showed that intestinal mucosal cholesterol and bile acid synthesis and metabolism were significantly affected. It was involved in the regulation of cholesterol, bile acid synthesis and metabolism, as well as cholesterol, bile acid secretion, absorption, transport and other physiological processes. The results showed that CyHV-2 disease had a serious effect on gene expression in intestinal mucosa and cholesterol. The synthesis and metabolism of bile acids and the gene expression of bile acid enterohepatic circulation pathway have a serious effect, which will lead to cholesterol in diseased fish. The content of bile acid in serum of fish with CyHV-2 disease decreased by 99%, and the content of cholesterol decreased by 10%, which confirmed the trend of differentially expressed genes.
