一個(gè)與煙草TMV抗性基因N緊密連鎖的共顯性SSR標(biāo)記

【摘要】:煙草普通花葉病(Tobacco Mosaic Virus,TMV)是煙葉生產(chǎn)的主要病害,為建立煙草抗TMV分子標(biāo)記輔助選擇體系,本研究以高抗TMV烤煙品種Coker176和易感TMV烤煙品種Y3以及它們的BC1F1群體為試材,采用分離集團(tuán)分組分析法(Bulked Segregation Analysis,BSA)和簡(jiǎn)單序列重復(fù)(Simple Sequence Repeat,SSR)標(biāo)記技術(shù),篩選與TMV抗性基因N緊密連鎖的共顯性SSR標(biāo)記。通過(guò)對(duì)18764對(duì)SSR引物的分析表明,有396對(duì)SSR引物在兩親本間有多態(tài),選出1對(duì)SSR引物TM508-007擴(kuò)增出的標(biāo)記與TMV抗性的N基因緊密連鎖,遺傳連鎖距離為0.41 c M,證實(shí)獲得的與TMV抗性基因N緊密連鎖的共顯性SSR標(biāo)記穩(wěn)定可靠,可用于煙草抗TMV分子標(biāo)記輔助選擇。
[Abstract]:Tobacco Mosaic virus (TMV) is a major disease in tobacco leaf production. It is a molecular marker-assisted selection system for tobacco resistance to TMV. In this study, high resistant TMV flue-cured tobacco varieties Coker176 and susceptible TMV varieties Y3 and their BC1F1 populations were used as the test materials. The separated Segregation Analysis was analyzed by grouping analysis of separated groups. BSA) and simple Sequence repeat SSRs. Screening codominant SSR markers closely linked to N of TMV resistance gene. The analysis of 18764 pairs of SSR primers showed that there were 396 pairs of SSR primers with polymorphism between the two parents. A pair of SSR primers TM508-007 was selected to amplify the marker closely linked to the N gene of TMV resistance, and the genetic linkage distance was 0.41 cm. It was proved that the codominant SSR markers closely linked to the TMV resistance gene N were stable and reliable and could be used in the assisted selection of tobacco TMV resistant molecular markers.
