腫瘤干細(xì)胞中受H3K9me2調(diào)控的干性基因的篩選

【摘要】:為了分析比較甲基轉(zhuǎn)移酶G9a和組蛋白H3K9me2修飾在膠質(zhì)瘤干細(xì)胞與非干細(xì)胞中存在的差異,篩選出維持膠質(zhì)瘤干細(xì)胞干性的相關(guān)基因。通過(guò)G9a抑制劑促進(jìn)U87細(xì)胞成球和過(guò)表達(dá)G9a促進(jìn)U87細(xì)胞分化的方法,培養(yǎng)了成球的膠質(zhì)瘤干細(xì)胞和貼壁的非干細(xì)胞,這兩種細(xì)胞的CD133表達(dá)差異明顯。再利用H3K9me2抗體通過(guò)Ch IP-seq技術(shù)比較H3K9me2修飾在干細(xì)胞組與非干細(xì)胞組中的差異,在存在差異的基因中,對(duì)TSS±2 000 bp范圍內(nèi)的基因進(jìn)行了GO分析,并隨機(jī)選出10個(gè)轉(zhuǎn)錄因子進(jìn)行QPCR驗(yàn)證,結(jié)果與Ch IP-seq實(shí)驗(yàn)基本一致。
[Abstract]:To compare the difference of methyltransferase G9a and histone H3K9me2 in glioma stem cells and non-stem cells. The genes related to maintaining stem cell dryness of glioma were screened. G9a inhibitor was used to promote the formation of U87 cells and overexpression of G9a to promote the differentiation of U87 cells. Glioma stem cells and adherent non-stem cells were cultured. H3K9me2 antibody was used to compare the difference of H3K9me2 modified H3K9me2 between stem cell group and non-stem cell group by Ch IP-seq technique. Among the differentially expressed genes, go was used to analyze the genes in the range of TSS 鹵2 000 BP, and 10 transcription factors were randomly selected for QPCR validation. The results were consistent with Ch IP-seq experiment.
