【摘要】：在構(gòu)建的角毛殼菌(Chaetomium cupreum)菌絲體cDNA文庫(kù)中獲得絲切蛋白(Cofilin)的ESTs序列片段,為研究角毛殼菌的生物特性,提高其生物防治效果,以角毛殼菌總RNA反轉(zhuǎn)錄產(chǎn)物為模板進(jìn)行PCR擴(kuò)增,成功獲得了Cofilin基因cDNA序列.生物信息學(xué)分析結(jié)果表明,該基因全長(zhǎng)468bp,編碼155個(gè)氨基酸,編碼蛋白理論分子量17kD,等電點(diǎn)是4.99,α-螺旋和無(wú)規(guī)則卷曲是其主要的二級(jí)結(jié)構(gòu),為親水性蛋白質(zhì).該蛋白有6個(gè)位點(diǎn)發(fā)生Ser磷酸化,4個(gè)位點(diǎn)發(fā)生Tyr磷酸化,有4個(gè)保守區(qū),ClustalX分析說(shuō)明該蛋白與柄孢霉(Podospora anserina)和球毛殼菌(C.g lobosum)親緣關(guān)系較近.
[Abstract]:The ESTs sequence fragment of Cofilin was obtained from the constructed cDNA library of Chaetomium cupreum. In order to study the biological characteristics of Trichoderma keratoderma and improve its biological control effect, the total RNA reverse transcription product was used as template for PCR amplification. The cDNA sequence of Cofilin gene was successfully obtained. Bioinformatics analysis showed that the full length of the gene was 468 BP, encoding 155 amino acids, and the theoretical molecular weight of encoding protein was 17 KD. The isoelectric point is 4.99, and 偽 -helix and irregular crimp are the main secondary structures, which are hydrophilic proteins. There are 6 sites of Ser phosphorylation and 4 sites of Tyr phosphorylation. There are four conservative areas. ClustalX analysis showed that the protein was closely related to Podospora anserina and C. G lobosum.
