人miR-17-92基因簇載體構(gòu)建及其在293T細(xì)胞中的表達(dá)驗(yàn)證

【摘要】:目的:研究人miR-17-92基因簇在腎癌等疾病發(fā)生過程中的功能。方法:采用基因重組技術(shù),將miR-17-92基因簇亞克隆至pc DNA4真核細(xì)胞表達(dá)載體,構(gòu)建miR-17-92基因簇真核表達(dá)載體;通過脂質(zhì)體轉(zhuǎn)染的方法將miR-17-92的過表達(dá)質(zhì)粒和對(duì)照質(zhì)粒分別轉(zhuǎn)染293T細(xì)胞,并用實(shí)時(shí)定量PCR技術(shù)驗(yàn)證轉(zhuǎn)染24和48 h后細(xì)胞內(nèi)pri-miR-17-92和miR-17、miR-18a、miR-19a、miR-19b1、miR-20a、miR-92a1等分子的相對(duì)過表達(dá)比例。結(jié)果:miR-17-92基因簇真核表達(dá)載體構(gòu)建成功,并在293T細(xì)胞中實(shí)現(xiàn)了顯著性過表達(dá)效果。結(jié)論:為進(jìn)一步研究人miR-17-92基因簇在腎臟發(fā)育過程中的功能及其與腎臟癌癥發(fā)病相關(guān)性的研究提供了實(shí)驗(yàn)基礎(chǔ)。
[Abstract]:Objective: to study the function of human miR-17-92 gene cluster in renal cell carcinoma and other diseases. Methods: gene recombination technique was used. The miR-17-92 gene cluster was subcloned into the eukaryotic expression vector of PC DNA4, and the eukaryotic expression vector of miR-17-92 gene cluster was constructed. The overexpression plasmids of miR-17-92 and control plasmids were transfected into 293T cells by liposome transfection. The intracellular pri-miR-17-92 and miR-17 miR-18a miR-19a were confirmed by real-time quantitative PCR after transfection for 24 and 48 hours. The relative overexpression ratio of miR-19b1mmiR-20ahmiR-92a1 and other molecules was compared. Results the eukaryotic expression vector of the gene cluster of miR-17-92 was successfully constructed. Significant overexpression was achieved in 293T cells. Conclusion:. It provides an experimental basis for the further study of the function of human miR-17-92 gene cluster in renal development and its association with renal cancer.
