土壤桿菌氧調(diào)控基因fnrN突變株發(fā)酵性能及基于RNA-Seq的基因表達(dá)

【摘要】:【目的】研究Crp家族轉(zhuǎn)錄調(diào)節(jié)因子fnr N突變對(duì)土壤桿菌ATCC 31749發(fā)酵性能和基因表達(dá)的影響。【方法】利用三親結(jié)合法將構(gòu)建的自殺式質(zhì)粒p JQ-fnr N-kan導(dǎo)入土壤桿菌ATCC31749中,從而獲得fnr N基因突變株(Δfnr N);分析Δfnr N的發(fā)酵特性;基于RNA-Seq對(duì)產(chǎn)膠期土壤桿菌ATCC 31749野生菌和Δfnr N差異表達(dá)基因進(jìn)行分析!窘Y(jié)果】fnr N的突變使土壤桿菌合成熱凝膠能力下降了22.0%,轉(zhuǎn)錄組分析發(fā)現(xiàn)在Δfnr N中186個(gè)基因表達(dá)發(fā)生顯著性變化(|log2(|fold change|)|≥1且q≤0.001),其中65%的基因表達(dá)上調(diào)。熱凝膠合成的關(guān)鍵基因crd ASC的表達(dá)受到不同程度的抑制,fnr N的突變使編碼σ因子的ecf R和編碼生物膜合成調(diào)節(jié)因子的sin R顯著下調(diào);Δfnr N菌中與細(xì)胞色素有關(guān)基因cyd AB、cy2、fix NOPQ的轉(zhuǎn)錄水平下調(diào)2-13倍!窘Y(jié)論】fnr N通過調(diào)控ecf R和sin R的表達(dá)調(diào)控?zé)崮z合成,通過調(diào)控fix NOPQ等基因的表達(dá)參與氧信號(hào)調(diào)控,該研究有助于豐富對(duì)土壤桿菌氧調(diào)控系統(tǒng)的認(rèn)識(shí)。
[Abstract]:[objective] to study the effect of Crp family transcription regulator fnr N mutation on ATCC of Agrobacterium tumefaciens. 31749 fermentation performance and gene expression. [methods] the constructed suicide plasmid p JQ-fnr N-kan was introduced into ATCC31749 of Agrobacterium tumefaciens. Thus, the mutant of fnr N gene (螖 fnr N) was obtained. The fermentation characteristics of 螖 fnr N were analyzed. The differentially expressed genes of ATCC 31749 wild bacteria and 螖 fnr N were analyzed based on RNA-Seq. [results] fnr. The mutation of N reduced the ability of Agrobacterium to synthesize thermal gel by 22.0%. Transcriptome analysis showed that 186 genes were significantly changed in 螖 fnr N (log2 (fold change) 鈮,
