AANAT基因編碼區(qū)多態(tài)性與家兔生長性狀的關(guān)聯(lián)分析

【摘要】:為研究五羥色胺-N-乙;D(zhuǎn)移酶(AANAT)基因編碼區(qū)(CDS區(qū))多態(tài)性和家兔生長性狀的關(guān)聯(lián)性,以天府黑兔、伊拉兔和新西蘭兔作為實(shí)驗(yàn)材料,對(duì)AANAT基因的外顯子進(jìn)行PCR擴(kuò)增,直接測序檢測家兔AANAT基因外顯子的單核苷酸多態(tài)性。結(jié)果表明:在3個(gè)兔品種的CDS區(qū)上均發(fā)現(xiàn)有5個(gè)完全連鎖突變位點(diǎn),分別是C.132TC、C.153TC、C.159 AT、C.234 AG、C.291 AG,形成了野生型TTTTAAAAAA、雜合型TCTCATAGAG、突變型CCCCTTGGGG 3種基因型,突變型新西蘭兔在56、70、84日齡的體重顯著高于野生型和雜合型(P0.05),而突變型伊拉兔和天府黑兔在70、84日齡的體重顯著高于雜合型和野生型(P0.05)。綜上可知,AANAT基因可作為肉兔生長性狀的候選基因。
[Abstract]:In order to study the association between polymorphism of serotonin N-acetyltransferase (AANATE) gene coding region and growth traits of rabbits, Tianfu black rabbit, Ira rabbit and New Zealand rabbit were used as experimental materials. The exon of AANAT gene was amplified by PCR. The single nucleotide polymorphisms of the exon of AANAT gene in rabbits were detected by direct sequencing. The results showed that there were five completely linked mutation sites in the CDS region of the three rabbits, namely C.132TC. The wild type TTTTAAAA, heterozygote TCTCATAGAG was formed. The body weight of mutant New Zealand rabbits was significantly higher than that of wild type and heterozygote (P0.05). However, the body weight of mutant Ira rabbits and Tianfu black rabbits was significantly higher than that of heterozygote and wild type P0.05N at 70 ~ 84 days old, which suggested that AANAT gene could be used as a candidate gene for growth traits of meat rabbits.
